蠶絲-PLGA網(wǎng)狀支架的力學(xué)強(qiáng)度檢測及其與骨髓間充質(zhì)干細(xì)胞共培養(yǎng)

【摘要】:目的進(jìn)行蠶絲-PLGA復(fù)合網(wǎng)狀支架的力學(xué)強(qiáng)度檢測及其與骨髓間充質(zhì)干細(xì)胞的體外共培養(yǎng)。方法采用緯編針織法制備蠶絲絲素-PLGA纖維復(fù)合網(wǎng)狀支架,并進(jìn)行力學(xué)強(qiáng)度測試。在網(wǎng)狀支架上滴加Ⅰ型膠原并冷凍干燥,再用纖維連接蛋白表面修飾后,種植兔骨髓間充質(zhì)干細(xì)胞進(jìn)行體外共培養(yǎng),觀察細(xì)胞與支架復(fù)合生長情況。結(jié)果蠶絲-PLGA網(wǎng)狀支架寬約5 mm。支架的最大載荷、抗拉強(qiáng)度、斷點伸長率、彈性模量分別為(118.32±11.21)N、(52.90±5.03)MPa、49.79%±5.16%、(177.25±19.18)MPa。支架-MSCs復(fù)合物培養(yǎng)2 d的掃描電鏡觀察顯示:MSCs細(xì)胞黏附于支架表面呈立體狀生長,細(xì)胞呈梭形,增殖良好,并分泌基質(zhì)。結(jié)論蠶絲-PLGA復(fù)合網(wǎng)狀支架具有良好的力學(xué)性能與細(xì)胞相容性,是一種較有潛力的組織工程韌帶的支架材料。
[Abstract]:Objective to investigate the mechanical strength of silk-PLGA composite reticular scaffold and co-culture with bone marrow mesenchymal stem cells in vitro. Methods the silk fibroin-PLGA fiber composite mesh scaffold was prepared by weft knitting method and its mechanical strength was tested. The mesenchymal stem cells of rabbit bone marrow were co-cultured with fibronectin after adding type I collagen and freeze-drying on the reticular scaffold. The growth of the cells and scaffolds was observed. Results the width of silk-PLGA mesh scaffold was about 5 mm. The maximum load, tensile strength, elongation at breakpoint and elastic modulus of the scaffold were (118.32 鹵11.21) N, (52.90 鹵5.03) MPA, (49.79% 鹵5.16) and (177.25 鹵19.18) MPA, respectively. SEM observation of scaff-MSCs complex culture for 2 days showed that the cells adhered to the scaffold were stereoscopic, the cells were fusiform, proliferated well, and secreted matrix. Conclusion Silkworm filament-PLGA composite reticular scaffold has good mechanical properties and cytocompatibility and is a potential scaffold for tissue engineering ligament.
