低強(qiáng)度脈沖超聲波對(duì)富血小板纖維蛋白釋放生長(zhǎng)因子的影響

【摘要】:目的:探討低強(qiáng)度脈沖超聲波(LIPUS)對(duì)富血小板纖維蛋白(PRF)釋放3種生長(zhǎng)因子(堿性成纖維細(xì)胞生長(zhǎng)因子(bFGF),轉(zhuǎn)化生長(zhǎng)因子p1(TGF-p1),表皮生長(zhǎng)因子(EGF))的影響,為今后更有效的應(yīng)用PRF提供實(shí)驗(yàn)依據(jù)和理論基礎(chǔ)。 方法:1.選擇10名不吸煙的健康成人(22-27歲)男性志愿者,抽靜脈血20ml加入無任何抗凝劑的采血管中,即刻以3000r/min離心10分鐘,獲得初級(jí)的PRF凝膠,再將其靜置于干燥消毒的容器內(nèi)10min,使其自然收縮并釋放其內(nèi)的血清,然后用LIPUS輻照PRF所釋放的滲出液。2.根據(jù)輻照時(shí)間分為3組:分別為輻照LIPUS15分鐘組(L-15min組)、輻照LIPUS30分鐘組(L-30mmin組)和空白對(duì)照組(Control組)。3.實(shí)驗(yàn)組和對(duì)照組采用酶聯(lián)免疫吸附法(ELISA)測(cè)定PRF滲出液中的3種生長(zhǎng)因子含量。所得數(shù)據(jù)用SPSS18.0軟件進(jìn)行統(tǒng)計(jì)學(xué)分析。 結(jié)果:低強(qiáng)度脈沖超聲波作用于富血小板纖維蛋白后3種生長(zhǎng)因子濃度變化:bFGF的L-15min組(318.60pg/ml)大于L-30min組(293.00pg/ml)和Control組(278.20pg/ml),3組之間差異均有顯著性意義(P0.05); TGF-β1的L-15min組(7259pg/ml)大于L-30min組(6035pg/ml)和Control組(5503pg/ml),3組之間差異均有顯著性意義(P0.05); EGF的Control組、L-15min組和L-30min組平均濃度分別為177.87pg/ml、181.80pg/ml和177.91pg/ml,3組之間濃度變化均無明顯差異(P0.05)。 結(jié)論:低強(qiáng)度脈沖超聲波可能使富血小板纖維蛋白所釋放的生長(zhǎng)因子的濃度增加。在臨床治療中,富血小板纖維蛋白聯(lián)合低強(qiáng)度脈沖超聲波,應(yīng)用于牙周組織再生修復(fù),能更有效的利用富血小板纖維蛋白。
[Abstract]:Objective: to investigate the effects of low intensity pulsed ultrasound (LIPUS) on the release of three growth factors (basic fibroblast growth factor (bFGF), transforming growth factor p1 (TGF-p1), epidermal growth factor (EGF) by platelet-rich fibrin (PRF). It provides experimental and theoretical basis for more effective application of PRF in the future. Method 1: 1. Ten healthy non-smoking male volunteers (22-27 years old) were enrolled in this study. 20ml was extracted from venous blood and added to the blood vessels without any anticoagulant. The primary gel was obtained by centrifugation with 3000r/min for 10 minutes. Then it was statically placed in a dry sterilized container for 10 mins to naturally contract and release the serum in it. Then LIPUS was used to irradiate the exudate of PRF. 2. According to the irradiation time, the rats were divided into three groups: irradiation LIPUS 15-minute group (L-15min group), irradiated LIPUS30-minute group (L-30mmin group) and blank control group (Control group) .3. Enzyme linked immunosorbent assay (Elisa) was used to determine the content of three growth factors in PRF exudate. The data were analyzed by SPSS 18.0 software. Results: there were significant differences between L-15min group (318.60pg/ml) and control group (278.20pg/ml) and L-15min group (7259pg/ml) of TGF- 尾 1 compared with L-30min group (P0.05) and L-15min group (7259pg/ml) with TGF- 尾 1 as compared with L-30min group (P 0.05). There were significant differences between 6035pg/ml and 5503pg/ml groups (P0.05), but there was no significant difference between the control group (177.87pgml / ml) and the control group (177.91pgml / ml) (P0.05). Conclusion: low intensity pulsed ultrasound may increase the concentration of growth factor released by platelet rich fibrin. In clinical treatment, platelet-rich fibrin combined with low-intensity pulsed ultrasound can be used in periodontal tissue regeneration and repair, and it can be used more effectively to make use of platelet-rich fibrin.
