嬰兒皮膚組織成纖維細(xì)胞系的建立及生物學(xué)特性

【摘要】:采集嬰兒皮膚組織,用組織塊法制備原代細(xì)胞,通過(guò)差速消化和差速貼壁法純化細(xì)胞,擴(kuò)大培養(yǎng)至第3代液氮凍存,復(fù)蘇細(xì)胞后進(jìn)行活力、形態(tài)、生長(zhǎng)曲線、微生物污染、核型、乳酸脫氫同工酶譜及熒光蛋白質(zhì)粒轉(zhuǎn)染表達(dá)等特性研究.結(jié)果顯示,嬰兒皮膚組織細(xì)胞呈成纖維型,生長(zhǎng)狀態(tài)良好;生長(zhǎng)曲線呈"S"型,最大增值濃度為1.64×105/mL,倍增時(shí)間為63.12 h;細(xì)菌、真菌、支原體檢測(cè)均為陰性;染色體2n=46,二倍體為86%,占主體;乳酸脫氫酶同工酶電泳圖譜顯示有5條譜帶;外源質(zhì)粒在該細(xì)胞中能進(jìn)行復(fù)制和表達(dá).表明已成功建立了嬰兒皮膚組織成纖維細(xì)胞系,這為復(fù)合人工皮膚的組織工程學(xué)研究奠定了前期工作基礎(chǔ),也為皮膚相關(guān)疾病的研究提供了實(shí)驗(yàn)條件,因而具有重要的理論和實(shí)踐意義.
[Abstract]:The skin tissues of infants were collected and primary cells were prepared by tissue block method. The cells were purified by differential digestion and differential adherence method. The cells were expanded to the third generation of liquid nitrogen cryopreservation. After resuscitation, the cells were resuscitated for vitality, morphology, growth curve and microbial contamination. Karyotype, lactate dehydrogenation isozyme and fluorescent protein transfection and expression were studied. The results showed that the skin tissue cells of infant had fibroblast type, and the growth curve was "S" type, the maximum increment concentration was 1.64 脳 10 ~ 5 / mL, the doubling time was 63.12 h, and the detection of bacteria, fungi and mycoplasma were all negative. Chromosome 2nnc46, diploid 86.Lactic dehydrogenase isozyme electrophoresis showed that there were five bands, and exogenous plasmids could be duplicated and expressed in the cell. The results indicated that the infantile skin tissue fibroblast cell line had been successfully established, which laid a foundation for the study of tissue engineering of composite artificial skin and provided experimental conditions for the study of skin related diseases. Therefore, it has important theoretical and practical significance.
