動(dòng)態(tài)多孔海綿結(jié)構(gòu)多層膜負(fù)載溶菌酶用于抗菌涂層的研究

【摘要】:以聚丙烯酸(PAA)和聚乙烯亞胺(PEI)為構(gòu)筑單元,運(yùn)用層層自組裝技術(shù)制備了聚電解質(zhì)多層膜.該多層膜具有獨(dú)特的動(dòng)態(tài)特點(diǎn)——經(jīng)酸處理后膜內(nèi)部形成海綿狀通孔結(jié)構(gòu),該海綿結(jié)構(gòu)在飽和水蒸氣的處理下,多孔結(jié)構(gòu)能夠閉合,重新回到致密的膜結(jié)構(gòu).借助該種動(dòng)態(tài)多層膜平臺(tái),能夠簡(jiǎn)單有效地通過(guò)毛細(xì)作用力將溶菌酶負(fù)載并固定于多層膜中,為制備基于抗菌蛋白的抗菌涂層提供了新的方法.掃描電鏡表征了多層膜動(dòng)態(tài)變化過(guò)程,激光共聚焦顯微鏡表征了溶菌酶在膜內(nèi)的分布情況,并測(cè)定了溶菌酶載入量及其釋放動(dòng)力學(xué).進(jìn)一步的抗菌測(cè)試表明該種抗菌涂層在溶菌酶和PEI的共同作用下可以有效地抑制金黃色葡萄球菌.將多層膜同時(shí)負(fù)載溶菌酶和乳鐵蛋白,提升了涂層對(duì)大腸桿菌的殺菌效果.
[Abstract]:Polyelectrolyte multilayer films were prepared by layer-by-layer self-assembly technique using polyacrylic acid (PAA) and polyethyleneimine (PEI) as building units. The multilayer membrane has unique dynamic characteristics: the porous structure is formed in the membrane after acid treatment, and the porous structure can be closed and returned to the dense membrane structure under saturated water vapor treatment. With the help of this dynamic multilayer film platform, lysozyme can be simply and effectively loaded and immobilized in the multilayer film by capillary force, which provides a new method for the preparation of antibacterial coating based on antibacterial protein. The dynamic process of the multilayer membrane was characterized by scanning electron microscope and the distribution of lysozyme in the membrane was characterized by laser confocal microscopy. The loading amount of lysozyme and its release kinetics were measured. Further antimicrobial tests showed that the coating could effectively inhibit Staphylococcus aureus under the combined action of lysozyme and PEI. The multilayer film was loaded with lysozyme and lactoferrin simultaneously to improve the bactericidal effect of the coating on Escherichia coli.
