【摘要】：背景:雖然當(dāng)前有多種方法可以觀察和檢測(cè)軟骨內(nèi)成骨早期血管新生,但是均存在一定的不足。目的:探討四環(huán)素和茜素絡(luò)合酮間接標(biāo)記軟骨內(nèi)成骨過(guò)程中新生血管的可能性。方法:制備新西蘭兔雙側(cè)橈骨骨缺損模型并植入β-磷酸三鈣材料,分別于術(shù)后第1天和第15天注射四環(huán)素和茜素絡(luò)合酮,第28天取材。部分標(biāo)本在墨汁灌注后行硬組織切片熒光/光學(xué)顯微鏡觀察,部分標(biāo)本經(jīng)脫鈣后行免疫組化染色觀察,比較軟骨內(nèi)成骨過(guò)程中親骨熒光素標(biāo)記管腔結(jié)構(gòu)與免疫組化、墨汁灌注標(biāo)記血管結(jié)構(gòu)的一致性。結(jié)果與結(jié)論:(1)親骨熒光素標(biāo)記的管腔結(jié)構(gòu)經(jīng)免疫組化染色證實(shí)為CD34陽(yáng)性的血管結(jié)構(gòu);(2)在熒光顯微鏡下,親骨熒光素標(biāo)記的血管形態(tài)與墨汁灌注的血管形態(tài)一致;熒光素標(biāo)記后經(jīng)墨汁灌注的血管在熒光顯微鏡下可見(jiàn)熒光管腔中有黑色墨汁走行;(3)此外,親骨熒光素標(biāo)記的管腔結(jié)構(gòu)顏色絢麗、三維結(jié)構(gòu)更加生動(dòng),可通過(guò)不同顏色的熒光顯示不同時(shí)期的血管新生和演變過(guò)程,具有獨(dú)特的優(yōu)勢(shì),可用于軟骨內(nèi)成骨早期血管發(fā)生的形態(tài)學(xué)檢測(cè)。
[Abstract]:Background: although there are many methods to observe and detect early angiogenesis in endochondral osteogenesis, there are some deficiencies. Aim: to investigate the possibility of indirect labeling of neovascularization with tetracycline and alizarin complex ketone during endochondral osteogenesis. Methods: the bilateral radial bone defect model of New Zealand rabbits was established and 尾 -tricalcium phosphate was implanted. Tetracycline and alizarin complex ketone were injected on the 1st and 15th day after operation, and the materials were obtained on the 28th day. Some of the specimens were observed by fluorescence / optical microscopy after infusing with Chinese ink, and others were observed by immunohistochemical staining after decalcification to compare the structure and immunohistochemistry of osteophile fluorescein labeling during endochondral osteogenesis. Consistency of blood vessel structure with ink perfusion labeling. Results and ConclusionThe vascular structure labeled with osteofluorescein was proved to be CD34 positive by immunohistochemical staining. Under fluorescence microscope, the morphology of the vessel labeled with osteofluorescein was consistent with that of the blood vessel infused with ink. Fluorescein labeled vessels infused with ink showed black ink in the fluorescent cavity under fluorescence microscope. In addition, the structure of the lumen labeled with osteofluorescein was colorful, and the three-dimensional structure was more vivid. It can display the process of angiogenesis and evolution at different stages by different color fluorescence, which has unique advantages and can be used to detect the morphology of angiogenesis in early stage of endochondral osteogenesis.
