新型鼻腔填塞材料聚乙二醇雙丙烯酸酯復合物水凝膠的細胞相容性

【摘要】:背景:前期研究發(fā)現(xiàn)聚乙二醇雙丙烯酸酯和殼聚糖形成的復合物可生物降解吸收,具有抗炎消腫能促進創(chuàng)面愈合作用,且生物相容性好,然而該復合物質地較脆,鑒于鼻腔解剖結構,需要一種具有力學支撐性能的材料來克服這種缺陷。目的:觀察新型可降解吸收鼻腔填塞材料聚乙二醇雙丙烯酸酯復合物水凝膠的細胞相容性。方法:通過光交聯(lián)將聚乙二醇雙丙烯酸酯、殼聚糖、聚乙烯醇材料復合,形成聚乙二醇雙丙烯酸酯復合物水凝膠,根據(jù)國際通行醫(yī)療器械生物學評價標準采用直接解觸法及間接浸提法評價其細胞相容性。(1)直接解觸法:將聚乙二醇雙丙烯酸酯復合物水凝膠與小鼠成纖維細胞L929共培養(yǎng)24 h,觀察形態(tài)變化;(2)間接浸提法:分別以100,50μL聚乙二醇雙丙烯酸酯復合物水凝膠浸提液及100μL RPMI1640培養(yǎng)液培養(yǎng)小鼠成纖維細胞L929,CCK-8法檢測培養(yǎng)1-3 d的增殖及細胞毒性。結果與結論:(1)直接解觸法:材料周圍細胞形態(tài)呈伴突起的梭形或星形的扁平狀結構,貼壁性生長,細胞飽滿且折光性強,整體呈生長趨勢,可見該材料生物相容性良好;(2)間接浸提法:隨著時間的增加,含復合物水凝膠浸提液組細胞數(shù)量增加,細胞相對增殖率逐漸增加,細胞毒性為0至1級;(3)結果表明:聚乙二醇雙丙烯酸酯復合物水凝膠材料具有良好的細胞相容性。
[Abstract]:Background: previous studies have shown that the complex formed by polyethylene glycol bisacrylates and chitosan can be biodegradable and absorbable, has anti-inflammatory and swelling effects, and has good biocompatibility, but the texture of the complex is brittle. In view of the anatomical structure of the nasal cavity, a mechanical support material is needed to overcome this defect. Aim: to observe the cytocompatibility of a new biodegradable nasal packing material polyethylene glycol diacrylate complex hydrogel. Methods: Polyethylene glycol bisacrylates, chitosan and polyvinyl alcohol were mixed by photocrosslinking to form polyglycol bisacrylates hydrogels. According to the international standard of biological evaluation of medical devices, direct desatalysis method and indirect extraction method were used to evaluate their cytocompatibility.) direct desaturation method: Polyethylene glycol diacrylate complex hydrogel and mouse fibroblast L929 were treated with polyglycol diacrylate complex hydrogel. After 24 hours of co-culture, morphological changes were observed. Indirect extraction method: the proliferation and cytotoxicity of mouse fibroblasts cultured in 100 渭 L polyethylene glycol diacrylate complex hydrogel solution and 100 渭 L RPMI1640 medium were detected by L929CCK-8 method for 1-3 days. Results and conclusion: the cell around the material showed fusiform or starlike flat structure with protuberance, adherent growth, full cells and strong refraction. It can be seen that the material has good biocompatibility and indirect extraction method: with the increase of time, the number of cells and the relative proliferation rate of the cells increased with the increase of time. The results of cytotoxicity from 0 to 1) showed that the hydrogels of polyethylene glycol diacrylate complex had good cytocompatibility.
