大鼠筋膜源性細(xì)胞與肌源性細(xì)胞成骨能力的對比研究

【摘要】:背景:自體骨髓來源的間充質(zhì)干細(xì)胞具有多分化潛能,能向成骨、成軟骨細(xì)胞分化,加速骨組織及軟骨組織的形成,但是成骨能力各不相同,哪種細(xì)胞的成骨能力更強(qiáng)有待進(jìn)一步研究。目的:比較大鼠筋膜源性細(xì)胞與肌源性細(xì)胞的成骨能力。方法:從20只大鼠骨骼肌筋膜分離出筋膜源性細(xì)胞,從骨骼肌肌腹中分離出肌源性細(xì)胞。用流式細(xì)胞儀分選筋膜源性細(xì)胞和肌源性細(xì)胞,培養(yǎng)細(xì)胞。利用retro-BMP4病毒轉(zhuǎn)染FDC-LacZ細(xì)胞。經(jīng)過2次轉(zhuǎn)染,轉(zhuǎn)染的筋膜源性細(xì)胞通過Lac Z和堿性磷酸酶的染色而測定出轉(zhuǎn)染效率。結(jié)果與結(jié)論:肌源性細(xì)胞產(chǎn)生的軟骨細(xì)胞、鈣鹽沉積均多于筋膜源性細(xì)胞。提示大鼠肌源性細(xì)胞的成骨能力強(qiáng)于筋膜源性細(xì)胞。
[Abstract]:Background: mesenchymal stem cells derived from autologous bone marrow have the potential to differentiate into osteoblasts, chondroblasts, and accelerate the formation of bone tissue and cartilage tissue, but the osteogenic capacity is different. Which cell has more osteogenic ability needs further study. Objective: to compare the osteogenic ability of rat fascia derived cells and myogenic cells. Methods: fascial-derived cells were isolated from 20 rat skeletal muscle fascia and myogenic cells were isolated from skeletal muscle belly. The fascia derived cells and myogenic cells were separated by flow cytometry and cultured. FDC-LacZ cells were transfected with retro-BMP4 virus. After twice transfection, the transfected fascia derived cells were stained with Lac Z and alkaline phosphatase to determine the transfection efficiency. Results and conclusion: the calcium deposition of chondrocytes produced by myogenic cells was more than that of fascial-derived cells. The results suggest that the osteogenic ability of rat myogenic cells is stronger than that of fascial-derived cells.
