軟骨細(xì)胞及骨髓間充質(zhì)干細(xì)胞共培養(yǎng)修復(fù)關(guān)節(jié)軟骨缺損

【摘要】:背景:關(guān)節(jié)軟骨損傷多由創(chuàng)傷及骨關(guān)節(jié)炎引起,其治療是臨床研究的難點(diǎn)和熱點(diǎn)。組織工程技術(shù)的發(fā)展為修復(fù)關(guān)節(jié)軟骨缺損提供了新方法,但軟骨細(xì)胞來源不足一直是一個(gè)難題。目的:探討軟骨細(xì)胞及骨髓間充質(zhì)干細(xì)胞共培養(yǎng)修復(fù)關(guān)節(jié)軟骨缺損的可行性,評(píng)價(jià)修復(fù)效果。方法:體外培養(yǎng)擴(kuò)增豬骨髓間充質(zhì)干細(xì)胞及關(guān)節(jié)軟骨細(xì)胞,共培養(yǎng)2代,接種于聚乳酸-聚羥基乙酸聚合物上繼續(xù)共培養(yǎng)2周,構(gòu)建共培養(yǎng)組織工程軟骨(共培養(yǎng)組);以單純軟骨細(xì)胞構(gòu)建組織工程軟骨為對(duì)照組,回植修復(fù)關(guān)節(jié)軟骨缺損;空白組軟骨缺損不作處理。半年后行大體觀察及組織學(xué)觀察,定量檢測(cè)Ⅱ型膠原染色面積及氨基糖胺聚糖含量。結(jié)果與結(jié)論:(1)共培養(yǎng)組的修復(fù)組織呈軟骨樣,表面光滑平坦,與周圍關(guān)節(jié)軟骨組織整合良好;對(duì)照組呈纖維組織修復(fù);而空白組則無修復(fù);(2)組織學(xué)觀察顯示,共培養(yǎng)組結(jié)構(gòu)致密,細(xì)胞外基質(zhì)分布更均勻,與周圍關(guān)節(jié)軟骨組織及軟骨下骨組織整合良好;(3)共培養(yǎng)組的Ⅱ型膠原染色面積及糖胺聚糖含量均優(yōu)于對(duì)照組及空白組,差異有顯著性意義(P0.05);(4)綜上,軟骨細(xì)胞和骨髓間充質(zhì)干細(xì)胞共培養(yǎng)構(gòu)建組織工程軟骨能有效修復(fù)關(guān)節(jié)軟骨缺損。
[Abstract]:Background: articular cartilage injury is caused by trauma and osteoarthritis. The development of tissue engineering provides a new method for repairing articular cartilage defects, but the shortage of chondrocytes is always a difficult problem. Aim: to investigate the feasibility of co-culture of chondrocytes and bone marrow mesenchymal stem cells (BMSCs) for repair of articular cartilage defects. Methods: porcine bone marrow mesenchymal stem cells and articular chondrocytes were cultured in vitro. Co-culture of tissue engineered cartilage (co-cultured group); using pure chondrocyte to construct tissue engineered cartilage as control group and replantation to repair articular cartilage defect; the blank group was not treated with cartilage defect.
