【摘要】：目的： 研究5-FU對體外培養(yǎng)的人良性膽管瘢痕成纖維細胞的作用。 方法： 1、人良性膽管瘢痕成纖維細胞的復蘇及傳代培養(yǎng)。 2、分別用濃度為0.78125g/1、1.5625g/1、3.125g/1、6.25g/1、12.5g/1的5-FU加入培養(yǎng)的人良性膽管瘢痕成纖維細胞,在光鏡下分組觀察5-FU干預人良性膽管瘢痕成纖維細胞24h、48h、72h成纖維細胞的生長形態(tài)變化,并通過MTT比色法檢測以上各組成纖維細胞增殖活性并計算細胞生長抑制率。選擇半數(shù)抑制濃度(50%Inhibitiory concentration, IC50)值作為干預濃度(6.25g/L)。 3、流式細胞儀檢測在最適干預濃度下5-FU干預人良性膽管瘢痕成纖維細胞的凋亡情況。 4、免疫熒光化學法測定人良性膽管瘢痕成纖維細胞在5-FU最適干預濃度作用后TGF-β1、a-SMA、Ⅰ型、Ⅲ型膠原蛋白表達的強度。 5、Western Blot方法檢測TGF-β1、a-SMA、Ⅰ型、Ⅲ型膠原蛋白的表達量。 結果： 1、5-FU干預人良性膽管瘢痕成纖維細胞72小時后,其抑制率為50%的濃度為6.25g/L。 2、5-FU干預膽管瘢痕成纖維細胞后,其生長曲線隨著時間的延長和藥物濃度的增大而受到明顯的抑制。 3、5-FU干預人良性膽管瘢痕成纖維細胞后凋亡明顯增加。 4、干預濃度(6.25g/L)5-FU作用于體外培養(yǎng)的人良性膽管瘢痕成纖維細胞,通過免疫熒光化學法和Western Blot方法測定成纖維細胞TGF-β1、a-SMA、Ⅰ型、Ⅲ型膠原蛋白表達均低于對照組。 結論： 1.5-FU可抑制人體膽管瘢痕成纖維細胞的生長。 2.5-FU通過促進細胞凋亡的方式,抑制了人體膽管瘢痕成纖維細胞的生長； 3.5-FU干預人體膽管瘢痕成纖維細胞后,TGF-β1、a-SMA、Ⅰ型膠原蛋白、Ⅲ型膠原蛋白表達明顯降低。 4.本實驗進一步揭示了5-FU治療人良性膽管瘢痕狹窄的作用機制,為今后開展5-FU局部用藥防治人良性膽管瘢痕狹窄提供實驗依據(jù)。
[Abstract]:Objective: To study the effect of 5-FU on human benign bile duct scar fibroblasts in vitro. Methods: 1. Resuscitation and subculture of human benign bile duct scar fibroblasts. 2. The cultured human benign bile duct scar fibroblasts were added with 0.78125g / 1 / 1.5625g / 1 5-FU 3.125g / 1 5-FU 6.25g / 1 / 1 and 12.5g / 1 respectively. The growth and morphological changes of human benign bile duct scar fibroblasts treated with 5-FU for 24 hours and 48 hours were observed under light microscope. MTT colorimetric assay was used to detect the proliferative activity of the fibroblasts and to calculate the cell growth inhibition rate.
