硅酸鹽生物活性玻璃碎片殺菌的機(jī)制研究

【摘要】:當(dāng)今,有眾多的新型抗菌藥物和材料被研究和應(yīng)用。比如:光照TiO2殺菌、金屬離子殺菌、季銨鹽等等,它們的抗菌方式都屬于納米顆粒抗菌模式。生物活性玻璃就是其中的一種并且占有重要的地位。其擁有良好的生物活性、相容性、抗菌性,因此,擁有巨大的研究?jī)r(jià)值和廣闊的應(yīng)用前景。 生物活性玻璃有42S,45S,S53P4、58S,77S等數(shù)種。而本文研究所選擇代表性的45S5做為研究對(duì)象。它浸入液體環(huán)境中后,其表面發(fā)生一系列反應(yīng),最后在表面形成一層與人體骨骼相似的羥基磷灰石層,與人體組織細(xì)胞緊密結(jié)合。 本文取45S5生物活性玻璃與大腸桿菌混合,進(jìn)行平板菌落計(jì)數(shù),測(cè)定其抗菌率,通過(guò)實(shí)驗(yàn)發(fā)現(xiàn)5min、15min、30min、60min四個(gè)時(shí)間段的抗菌率分別為54.55%、58.72%、42.78%、55.95%,具有明顯殺菌效果。然后用透射電鏡觀察其殺菌過(guò)程,發(fā)現(xiàn)45S5及其表面碎片與細(xì)菌細(xì)胞接觸后對(duì)膜結(jié)構(gòu)產(chǎn)生破壞作用,導(dǎo)致細(xì)菌死亡;或者45S5碎片進(jìn)入細(xì)菌細(xì)胞內(nèi)與細(xì)胞內(nèi)物質(zhì)作用,導(dǎo)致細(xì)菌死亡;通過(guò)觀察還發(fā)現(xiàn)兩者有同時(shí)作用的現(xiàn)象。
[Abstract]:Nowadays, many new antimicrobial agents and materials have been studied and applied. For example: illumination TiO2 sterilization, metal ion sterilization, quaternary ammonium salt and so on, their antimicrobial methods belong to nanoparticles antibacterial mode. Bioactive glass is one of them and plays an important role. Because of its good bioactivity, compatibility and antibacterial properties, it has great research value and wide application prospect. There are several kinds of bioactive glass, such as 42SU 45SN S53P4C58S77S and so on. The representative 45S5 is chosen as the research object in this paper. After it was immersed in liquid environment, a series of reactions took place on its surface. Finally, a hydroxyapatite layer similar to human skeleton was formed on the surface, which combined closely with human tissue and cells. In this paper, 45S5 bioactive glass was mixed with Escherichia coli, and its antibacterial rate was determined by plate colony count. It was found that the antibacterial rate was 54.55% and 58.72% in four periods of 5 min ~ 15 min ~ 30 min ~ 60 min ~ 60 min, respectively, and the bactericidal effect was obvious. Then the bactericidal process of 45S5 and its surface fragments were observed by transmission electron microscopy. It was found that 45S5 and its surface fragments had a destructive effect on the membrane structure after contact with bacterial cells, leading to the death of bacteria, or that 45S5 fragments enter bacteria cells and interact with intracellular substances. Causes the bacteria to die; through the observation also discovered the both to have simultaneously the phenomenon.
