皮膚組織工程新型材料的合成及表面改性的生物學(xué)基礎(chǔ)研究

【摘要】:PLGA是一種無毒可降解的高分子有機(jī)聚合物,其在機(jī)體內(nèi)代謝首先通過聚酯水解為乳酸和羥基乙酸,隨后被完全分解成為二氧化碳和水排出體外,生物相容性好。PVA水凝作為人工敷料的優(yōu)點(diǎn)是含水量豐富,不但可以保持創(chuàng)面濕潤,還具有吸水性可以吸收組織滲出液,同時其內(nèi)部大量貫通的孔隙有利于營養(yǎng)物質(zhì)和代謝廢物的運(yùn)輸,且又鎮(zhèn)痛作用,因而廣泛應(yīng)用與組織工程領(lǐng)域。本研究綜合了兩者的優(yōu)勢,并采用物理交聯(lián)的方式將二者復(fù)合,構(gòu)建一種類似皮膚結(jié)構(gòu)的雙層人工支架,使之更適合于臨床骨皮膚損傷、修復(fù)的需要。 明膠提取自動物的骨頭或結(jié)締組織,無毒無害,被廣泛用于食品、制藥及組織工程領(lǐng)域。在皮膚組織工程領(lǐng)域研究中,明膠除了具有促進(jìn)細(xì)胞粘附和生長的作用,還可以作為一種底物基質(zhì)與細(xì)胞外基質(zhì)產(chǎn)生競爭作用,防止傷口部位的蛋白酶消化細(xì)胞外基質(zhì)。明膠分子結(jié)構(gòu)上含有大量的羥基及少量的羧基和氨基,具有極強(qiáng)的親水性。因此,明膠分子中的氨基可以與光活性疊氮基團(tuán)發(fā)生縮合反應(yīng),形成酯類。本研究為進(jìn)一步提高人工皮膚支架材料的生物活性,以組織工程生物材料為基底,探討光固定表面改性的對其生物相容性的影響。 結(jié)果顯示:本研究利用光活性疊氮基團(tuán)與明膠分子的羧基發(fā)生了縮合反應(yīng),形成具有光活性的新的化合物—Az-G光活性明膠;通過在材料表面的固定效果和細(xì)胞學(xué)評價,證明Az-G光活性明膠可以穩(wěn)定的固定在生物材料表面,不會脫落,且細(xì)胞相容性好,有利于細(xì)胞粘附、生長和增殖;本研究首次將靜電紡絲膜和PVA水凝膠通過物理冷凍交聯(lián)的方法結(jié)合,,其機(jī)型性能和保水性遠(yuǎn)優(yōu)于單純的PLGA電紡絲膜或PVA水凝膠;采用模具法制備的“多孔”PVA水凝膠有利于細(xì)胞的粘附和生長。
[Abstract]:PLGA is a nontoxic and degradable polymeric organic polymer. Its metabolism is first hydrolyzed into lactic acid and glycolic acid by polyester and then completely decomposed into carbon dioxide and water.Good biocompatibility. PVA hydration as artificial dressing has the advantage of abundant water content, which not only keeps the wound moist, but also absorbs the tissue exudate.At the same time, a large number of perforated pores are conducive to the transport of nutrients and metabolic waste, and analgesic effect, so they are widely used in the field of tissue engineering.In this study, the advantages of both were synthesised, and they were combined by physical crosslinking to construct a kind of double-layer artificial scaffold similar to skin structure to make it more suitable for clinical bone skin injury and repair.Gelatin is extracted from animal bone or connective tissue. It is nontoxic and harmless. It is widely used in food, pharmaceutical and tissue engineering fields.In the field of skin tissue engineering, gelatin can not only promote cell adhesion and growth, but also act as a substrate to compete with extracellular matrix and prevent protease from digesting extracellular matrix.Gelatin contains a large number of hydroxyl groups, a small amount of carboxyl and amino groups, and has a strong hydrophilicity.Therefore, amino groups in gelatin molecules can be condensed with photoactive azide groups to form esters.In order to further improve the bioactivity of artificial skin scaffolds, the effect of surface modification on biocompatibility of tissue engineering biomaterials was investigated.The results showed that the photoactive azide group reacted with the carboxyl group of gelatin molecule to form a new photoactive compound -Az-G photoactive gelatin.The results showed that Az-G photoactive gelatin could be stably immobilized on the surface of biomaterials without falling off, and had good cell compatibility, which was beneficial to cell adhesion, growth and proliferation.In this study, the electrospun film and PVA hydrogel were combined by physical freezing crosslinking for the first time. The performance and water retention of the electrospun membrane and PVA hydrogel were much better than those of PLGA electrospinning film or PVA hydrogel.The "porous" PVA hydrogel prepared by mould method is beneficial to cell adhesion and growth.
