氧化鋯氧化鋁陶瓷復(fù)合材料對人牙周膜細(xì)胞增殖、凋亡和骨誘導(dǎo)活性的影響

【摘要】:背景:氧化鋯氧化鋁陶瓷復(fù)合材料具有優(yōu)良的抗腐蝕性、較高的強(qiáng)度和韌性,具有廣泛的應(yīng)用前景。目的:探究氧化鋯氧化鋁陶瓷復(fù)合材料對人牙周膜細(xì)胞增殖、凋亡和骨誘導(dǎo)活性的影響。方法:將第3-5代人牙周膜細(xì)胞分2組培養(yǎng),對照組細(xì)胞常規(guī)培養(yǎng),實(shí)驗(yàn)組與氧化鋯氧化鋁陶瓷復(fù)合材料共培養(yǎng),培養(yǎng)1-5 d,MTT法檢測細(xì)胞增殖,Tunel法檢測細(xì)胞凋亡,免疫組織化學(xué)法檢測細(xì)胞Ⅰ型膠原蛋白表達(dá),堿性磷酸酶染色檢測細(xì)胞成骨活性。結(jié)果與結(jié)論:培養(yǎng)1-5 d內(nèi),兩組細(xì)胞增殖、凋亡、堿性磷酸酶活性比較差異均無顯著性意義;培養(yǎng)5 d時(shí),兩組Ⅰ型膠原表達(dá)比較差異無顯著性意義;結(jié)果表明,氧化鋯氧化鋁陶瓷復(fù)合材料對牙周膜細(xì)胞增殖、凋亡和骨誘導(dǎo)活性均無影響。
[Abstract]:Background: zirconia ceramic composites have good corrosion resistance, high strength and toughness.Aim: to investigate the effects of zirconia ceramic composite on proliferation, apoptosis and bone induction activity of human periodontal ligament cells.Methods: the 3-5 passage human periodontal ligament cells were cultured in two groups, the control group was cultured routinely, the experimental group was co-cultured with zirconia ceramic composite material, and the cell proliferation was detected by MTT method for 1-5 days. The apoptosis was detected by Tunel method.The expression of type I collagen was detected by immunohistochemistry and osteogenesis activity was detected by alkaline phosphatase staining.Results and conclusion: there was no significant difference in cell proliferation, apoptosis and alkaline phosphatase activity between the two groups within 1-5 days, and there was no significant difference in the expression of type 鈪,
