骨髓間充質(zhì)干細(xì)胞復(fù)合Bio-oss修復(fù)兔顱骨缺損的組織學(xué)變化

【摘要】:背景:目前有一些關(guān)于骨髓間充質(zhì)干細(xì)胞復(fù)合異體骨、人工合成骨替代材料促進(jìn)骨缺損修復(fù)的研究,但尚未見骨髓間充質(zhì)干細(xì)胞復(fù)合Bio-oss用于兔顱骨缺損修復(fù)的報道。目的:觀察骨髓間充質(zhì)干細(xì)胞復(fù)合Bio-oss修復(fù)兔顱骨缺損的效果。方法:分離培養(yǎng)雄性兔骨髓間充質(zhì)干細(xì)胞,將其作為種子細(xì)胞,在雌兔顱骨采用外直徑6 mm的環(huán)形骨鉆制造全層骨缺損3個。將96只雌性兔隨機(jī)等分為4組,復(fù)合物組植入Bio-oss復(fù)合骨髓間充質(zhì)干細(xì)胞,Bio-oss組單純植入Bio-oss,骨髓間充質(zhì)干細(xì)胞組單純植入骨髓間充質(zhì)干細(xì)胞,空白組不進(jìn)行植入修復(fù)。每個組表面均覆蓋引導(dǎo)組織再生膜。結(jié)果與結(jié)論:復(fù)合物組成骨效果比其他3組更佳,Bio-oss組成骨效果比骨髓間充質(zhì)干細(xì)胞組和空白組更好,骨髓間充質(zhì)干細(xì)胞組與空白組成骨效果接近。說明以骨髓間充質(zhì)干細(xì)胞作為種子細(xì)胞,Bio-oss作為支架材料復(fù)合植入兔顱骨缺損區(qū)有明顯的促進(jìn)成骨作用,為臨床治療骨缺損提供新思路。
[Abstract]:Background: there have been some studies on bone marrow mesenchymal stem cells (BMSCs) combined with allogeneic bone, synthetic bone substitute materials to promote bone defect repair, but no reports of bone marrow mesenchymal stem cells (BMSCs) combined with Bio-oss for repairing rabbit skull defects have been reported.Objective: to observe the effect of bone marrow mesenchymal stem cells (BMSCs) combined with Bio-oss in repairing rabbit skull defect.Methods: male rabbit bone marrow mesenchymal stem cells were isolated and cultured as seed cells.96 female rabbits were randomly divided into 4 groups. The composite group was implanted with Bio-oss combined with bone marrow mesenchymal stem cells (BMSCs), the bone marrow mesenchymal stem cells (BMSCs) group was implanted with BMSCs only, and the blank group was not implanted with BMSCs.The surface of each group was covered with guided tissue regeneration membrane.Results and conclusion: the effect of composite bone composition was better than that of the other three groups. The effect of Bio-oss was better than that of bone marrow mesenchymal stem cells group and blank group. The effect of bone marrow mesenchymal stem cell group was similar to that of blank bone.It is concluded that the composite implantation of bone marrow mesenchymal stem cells (BMSCs) as seed cells and Bio-oss as scaffold material can obviously promote osteogenesis in rabbit skull defect and provide a new idea for clinical treatment of bone defect.
