【摘要】：目的觀察多聚甲醛固定對大鼠椎骨黏彈性力學(xué)性能的影響,從生物力學(xué)角度尋找松質(zhì)骨樣品的最佳保存方式。方法選擇8周齡健康雌性SD大鼠20只,手術(shù)分離獲得完整的腰椎L4和L5共40只,隨機(jī)平均分為固定組和對照組,固定組置于4%多聚甲醛中固定72 h,對照組置于5 m L EP管內(nèi)-20℃冷凍保存。兩組隨機(jī)各取10只分別行應(yīng)力松弛和蠕變實驗,7 200 s后收集樣品并通過顯微CT分析其微觀結(jié)構(gòu)變化。結(jié)果固定組松弛蠕變曲線相比對照組更加平滑,達(dá)到穩(wěn)態(tài)的時間較短,且500 s和7 200 s的松弛蠕變總量明顯降低(P0.01)。顯微CT結(jié)果顯示松弛蠕變實驗會引起骨小梁斷裂,且固定組比對照組骨小梁破壞更加嚴(yán)重。結(jié)論多聚甲醛固定法顯著降低大鼠椎骨的黏彈性,在力學(xué)負(fù)荷下更易造成其微觀結(jié)構(gòu)破壞,不利于松質(zhì)骨保存。
[Abstract]:Objective to observe the effect of paraformaldehyde fixation on the viscoelastic properties of vertebrae in rats and to find out the best preservation method of cancellous bone samples from the biomechanical point of view. Methods 20 healthy female SD rats aged 8 weeks were selected. A total of 40 lumbar vertebrae L4 and L5 were isolated and randomly divided into two groups: the fixed group and the control group.
