【摘要】:背景:殼聚糖生物材料能夠誘導(dǎo)骨髓間充質(zhì)干細(xì)胞向神經(jīng)細(xì)胞分化,殼聚糖衍生物羧甲基殼聚糖是否也能夠誘導(dǎo)骨髓間充質(zhì)干細(xì)胞向神經(jīng)細(xì)胞分化尚不清楚。目的:探討羧甲基殼聚糖溫敏凝膠對(duì)骨髓間充質(zhì)干細(xì)胞向神經(jīng)細(xì)胞分化的影響及其可能的機(jī)制。方法:取傳至3代的大鼠骨髓間充質(zhì)干細(xì)胞用不同質(zhì)量濃度(0,50,100,150,200,500 g/L)羧甲基殼聚糖溫敏凝膠浸提液培養(yǎng),對(duì)照組不添加羧甲基殼聚糖溫敏凝膠浸提液。MTT法檢測(cè)不同質(zhì)量濃度羧甲基殼聚糖溫敏凝膠浸提液對(duì)骨髓間充質(zhì)干細(xì)胞增殖的影響,Western blot檢測(cè)150 g/L羧甲基殼聚糖溫敏凝膠浸提液對(duì)骨髓間充質(zhì)干細(xì)胞NSE、Nestin、GAFP、Vimentin、NF-M、MAP-2、β3-tubulin以及Notch通路信號(hào)分子Notch1,Jag1表達(dá)水平的影響。結(jié)果與結(jié)論:(1)羧甲基殼聚糖溫敏凝膠浸提液促進(jìn)骨髓間充質(zhì)干細(xì)胞增殖,在質(zhì)量濃度為150 g/L時(shí),增殖率達(dá)到最高水平;(2)150 g/L羧甲基殼聚糖溫敏凝膠組NSE、Nestin、GAFP、Vimentin、NF-M、MAP-2、β3-tubulin的蛋白表達(dá)水平明顯高于對(duì)照組,表明羧甲基殼聚糖溫敏凝膠能夠誘導(dǎo)骨髓間充質(zhì)干細(xì)胞向神經(jīng)細(xì)胞分化;(3)羧甲基殼聚糖溫敏凝膠組Notch1和Jag1的蛋白表達(dá)水平明顯低于對(duì)照組,表明其在誘導(dǎo)分化過(guò)程中能抑制Notch信號(hào)通路的活性。
[Abstract]:Background: chitosan biomaterials can induce bone marrow mesenchymal stem cells to differentiate into neural cells. It is not clear whether chitosan derivative carboxymethyl chitosan can also induce the differentiation of bone marrow mesenchymal stem cells into neural cells. Objective: to investigate the differentiation of bone marrow mesenchymal stem cells into neural cells by carboxymethyl chitosan thermosensitive gel. Methods: rat bone marrow mesenchymal stem cells (BMSCs) were cultured with different concentrations of different concentrations of 0 50g / 100g / L carboxymethyl chitosan thermosensitive gel. Effect of different concentration of carboxymethyl chitosan thermo-sensitive gel extract on proliferation of bone marrow mesenchymal stem cells in control group. The effects of gel extract on the expression levels of NSE-Nestin-GAFP- VimentininCon NF-MfMAP-2, 尾 _ 3-tubulin and Notch signaling molecule Notch1- Jag1. Results and conclusion the proliferation of bone marrow mesenchymal stem cells was promoted by the solution of the thermally sensitive gel extract of carboxymethyl chitosan, which was isolated from the bone marrow mesenchymal stem cells. When the mass concentration was 150 g / L, the protein expression level of NSE Nestinine GAFP Vimentinn (NF-MMAP-2), 尾 3-tubulin (尾 3-tubulin) group was significantly higher than that of the control group (P < 0.05), and the proliferation rate reached the highest level at a concentration of 150 g 路L ~ (-1) / L carboxymethyl chitosan temperature-sensitive gel group. The results showed that carboxymethyl chitosan thermosensitive gel could induce bone marrow mesenchymal stem cells to differentiate into neural cells. The protein expression levels of Notch1 and Jag1 in carboxymethyl chitosan temperature-sensitive gel group were significantly lower than those in control group. These results suggest that it can inhibit the activity of Notch signaling pathway during differentiation induction.
