硅酸鹽生物活性玻璃與細(xì)菌吸附機(jī)制的研究

【摘要】:目前對于硅酸鹽生物玻璃的研究已經(jīng)有了一定的進(jìn)展,但是對于其抗菌作用的機(jī)制尚未形成系統(tǒng)的理論,尤其是對其與細(xì)菌吸附機(jī)制的研究目前在國內(nèi)還沒有文獻(xiàn)系統(tǒng)報(bào)告過,相關(guān)文獻(xiàn)表明,45S5抗菌原理為通過接觸性破壞細(xì)菌的細(xì)胞壁,導(dǎo)致細(xì)菌死亡,是對組織細(xì)胞無損害的特異性殺菌機(jī)制。因此研究其吸附機(jī)制將可能成為其抗菌實(shí)際應(yīng)用的關(guān)鍵理論基礎(chǔ),這對硅酸鹽生物活性玻璃的研究應(yīng)用有十分重大的意義,對與其它材料學(xué)及細(xì)胞或組織學(xué)的研究起到重要的參考作用。而本課題就是針對以上這些問題進(jìn)行探討,探究其抗菌機(jī)制下細(xì)菌吸附機(jī)制的作用模式,為抗菌課題進(jìn)一步研究提供理論基礎(chǔ),對其實(shí)際應(yīng)用提供理論支撐。 本課題主要通過文獻(xiàn)資料法、實(shí)驗(yàn)研究法、數(shù)理統(tǒng)計(jì)法觀察并探究金黃色葡萄球菌在45S5生物活性玻璃表面的吸附現(xiàn)象,主要通過探究生物活性材料45S5對金葡菌的吸附機(jī)理以及金葡菌自身的吸附力的形成兩方面來形成系統(tǒng)的理論。實(shí)驗(yàn)方法主要包括在體外采用細(xì)菌動(dòng)態(tài)吸附實(shí)驗(yàn),通過環(huán)境掃描電子顯微鏡、場發(fā)射電子顯微鏡等微觀實(shí)驗(yàn)手段探究其動(dòng)態(tài)變化,通過革蘭氏染色、96孔板生物膜檢測、ESS光密度曲線測定、等溫吸附曲線測定、45S5的電荷吸附方式等實(shí)驗(yàn)手段探究生物玻璃與細(xì)菌吸附的本質(zhì)機(jī)理。研究的主要結(jié)果如下: 1.45S5粉體對細(xì)菌有明顯吸附作用,粒徑小、濃度高的粉體吸附率較高。 2.45S5粉體與細(xì)菌的吸附過程是快速物理吸附過程,其吸附等溫線可用Langmiur吸附方程描述。其吸附方式為典型的雙電荷層吸附,電荷吸引力為其吸附的主要?jiǎng)恿Α?5S5粉體能影響細(xì)菌自身生物膜的形成,使其被阻斷于可逆性吸附階段,細(xì)菌依靠吸附于粉體上的方式大多為細(xì)菌的沉積、Brown運(yùn)動(dòng)、細(xì)菌表面與蛋白吸附層(CB)之間的靜電吸引、液體流動(dòng)等非特異性的物理因素。
[Abstract]:At present, some progress has been made in the study of silicate bioglass, but the mechanism of its antibacterial action has not yet formed a systematic theory, especially the mechanism of its adsorption with bacteria has not been systematically reported in domestic literature. The related literature shows that the antibacterial principle of PER45S5 is to destroy the cell wall of bacteria by contact, resulting in the death of bacteria. It is a specific bactericidal mechanism without damage to tissue and cell. Therefore, the study of its adsorption mechanism may become the key theoretical basis for its practical application, which is of great significance to the research and application of silicate bioactive glass. It plays an important reference role in the study of other materials and cell or histology. This subject is to discuss the above problems and explore the mechanism of bacterial adsorption under the antibacterial mechanism. It provides a theoretical basis for further research on antimicrobial subject and theoretical support for its practical application. In this paper, the adsorption of Staphylococcus aureus on the surface of 45S5 bioactive glass was observed and explored by the methods of literature, experiment and statistics. The mechanism of adsorption of bioactive material 45S5 to Staphylococcus aureus and the formation of adsorption capacity of Staphylococcus aureus were studied to form a systematic theory. The dynamic changes of ESS were investigated by means of environmental scanning electron microscope and field emission electron microscope, and the optical density curve of ESS was detected by Gram staining and 96 hole biofilm. The isothermal adsorption curve was used to determine the charge-adsorption mode of 45S5. The main results were as follows: (1) the intrinsic mechanism of biosorption between bioglass and bacteria was investigated. 1.45S5 powder has obvious adsorption effect on bacteria, and the adsorption rate of the powder with small particle size and high concentration is higher. The adsorption process of 2.45S5 powder and bacteria is a rapid physical adsorption process, and its adsorption isotherm can be described by Langmiur adsorption equation. The charge attraction is the main force of adsorption. 45S5 powder can affect the formation of bacterial biofilm and block it at the reversible adsorption stage. Non-specific physical factors such as electrostatic attraction and liquid flow between bacterial surface and protein adsorption layer (CBB).
