三維打印制備不同大孔形態(tài)含鍶介孔生物玻璃支架的體外細(xì)胞實(shí)驗(yàn)

【摘要】:背景:三維打印支架的大孔孔徑形態(tài)對(duì)生物組織工程支架理化性能及生物性能有很大的影響。目的:利用三維打印技術(shù)制備不同大孔結(jié)構(gòu)的含鍶介孔生物玻璃支架,探討其對(duì)MC3T3-E1細(xì)胞增殖和成骨分化的影響,尋找最佳的大孔形態(tài)。方法:利用三維打印技術(shù)制備含鍶介孔生物玻璃/聚己內(nèi)酯復(fù)合支架,其中縱橫交錯(cuò)的各纖維狀經(jīng)緯線之間的夾角設(shè)置為45°,60°和90°,根據(jù)其角度命名為45°,60°,90°含鍶介孔生物玻璃支架,檢測(cè)MC3T3-E1細(xì)胞在復(fù)合支架上的增殖能力和堿性磷酸酶活性。結(jié)果與結(jié)論:(1)CCK-8方法檢測(cè)結(jié)果表明,3組支架材料都能夠支持細(xì)胞增殖,第1天和第4天45°含鍶介孔生物玻璃支架上的細(xì)胞數(shù)量略高于60°和90°含鍶介孔生物玻璃支架,但差異無(wú)顯著性意義(P0.05);第7天45°含鍶介孔生物玻璃支架上的細(xì)胞數(shù)量顯著高于60°和90°含鍶介孔生物玻璃支架,差異有顯著性意義(P0.05);(2)在第14天,45°含鍶介孔生物玻璃支架的細(xì)胞堿性磷酸酶活性顯著高于60°和90°含鍶介孔生物玻璃支架,差異有顯著性意意義(P0.05);在第21天,3組支架的堿性磷酸酶活性差異無(wú)顯著性意義(P0.05);(3)結(jié)果表明,45°含鍶介孔生物玻璃支架相較于60°和90°含鍶介孔生物玻璃支架,更能促進(jìn)MC3T3-E1細(xì)胞的增殖和成骨分化。
[Abstract]:Background: the macroporous morphology of 3D printing scaffolds has great influence on the physical and chemical properties and biological properties of biomedical scaffolds. Objective: to prepare different macroporous mesoporous bioglass scaffolds with different macroporous structures by using 3D printing technology. In order to find the best macroporous morphology, the mesoporous glass / polycaprolactone composite scaffold containing strontium was prepared by three dimensional printing technique. Among them, the angles between the longitude and theodolite lines are set to 45 擄擄60 擄and 90 擄. According to their angles, they are named as 45 擄~ 60 擄~ 90 擄strontium containing mesoporous bioglass scaffolds, The proliferation and alkaline phosphatase activity of MC3T3-E1 cells on the composite scaffold were measured. Results and conclusions the results of CCK-8 assay showed that all three groups of scaffolds could support cell proliferation. On day 1 and day 4, the number of cells on 45 擄strontium containing mesoporous glass scaffolds was slightly higher than that of 60 擄and 90 擄strontium containing mesoporous glass scaffolds. On the 7th day, the number of cells on 45 擄strontium containing mesoporous glass scaffolds was significantly higher than that on 60 擄and 90 擄strontium containing mesoporous glass scaffolds. The cell alkaline phosphatase activity of 45 擄strontium containing mesoporous glass scaffolds was significantly higher than that of 60 擄and 90 擄strontium containing mesoporous bioglass scaffolds on the 14th day. On the 21st day, there was no significant difference in alkaline phosphatase activity among the three scaffolds. The results showed that 45 擄strontium containing mesoporous glass scaffolds were compared with 60 擄and 90 擄strontium containing mesoporous glass scaffolds. It can promote the proliferation and osteogenic differentiation of MC3T3-E1 cells.
