【摘要】:基因芯片技術日臻完善,但芯片探針的特異性與靈敏度之間的矛盾仍然是基因芯片在分子生物學和生物醫(yī)學方面應用研究的突出問題。使用肽核酸(PNA)做為芯片探針有望解決上述的問題。本文致力于基于光導向法的PNA芯片原位合成系統(tǒng)的設計與實現(xiàn),旨在開發(fā)用于合成中、高密度PNA芯片的自動化設備。 本文重點闡述了PNA芯片原位合成系統(tǒng)的設計與組裝,系統(tǒng)包括:虛擬掩模光刻系統(tǒng)、自動加樣系統(tǒng)及反應池裝置。研究內(nèi)容包括:在原有的虛擬掩模光刻系統(tǒng)的基礎上開發(fā)曝光控制軟件,通過控制數(shù)字微鏡器件(DMD)的圖像輸出,實現(xiàn)空間準確定位的多重光脫保護反應;設計以可編程邏輯控制器(PLC)為控制核心的自動加樣系統(tǒng),通過氣源動力裝置、預活化裝置及多向流體閥裝置,為固相PNA合成過程提供試劑的輸送、混合及洗滌等操作。本PNA原位合成系統(tǒng)在軟件指令的控制下,通過循序若干次的光脫保護反應、單體預活化處理、偶聯(lián)反應及蓋帽反應,實現(xiàn)基于光導向法的PNA芯片的原位合成。 本合成系統(tǒng)基本滿足中、高密度PNA芯片的合成需求,為PNA芯片的推廣及應用打下基礎。
[Abstract]:Gene chip technology is improving, However, the contradiction between the specificity and sensitivity of microarray probes is still a prominent problem in the application of gene chips in molecular biology and biomedicine. The use of peptide nucleic acid (PNA) as chip probes is expected to solve the above problems. This paper is devoted to the design and implementation of PNA chip in situ synthesis system based on optical guidance method. Designed to develop automation equipment for synthetic, high-density PNA chips. This paper focuses on the design and assembly of PNA chip in situ synthesis system. The system includes: virtual mask lithography system. The research contents include: developing exposure control software on the basis of the original virtual mask lithography system, and controlling the image output of the digital micromirror device (DMD). The automatic sampling system with PLC as the control core is designed, and the automatic sampling system is designed through the pneumatic power device, preactivation device and multidirectional fluid valve device. This PNA in-situ synthesis system, controlled by software instruction, is controlled by several successive optical deprotection reactions, monomer preactivation, coupling reactions and capping reactions. In-situ synthesis of PNA chip based on optical guidance method is realized. This synthesis system basically meets the synthesis demand of high density PNA chip, which lays a foundation for the popularization and application of PNA chip.
