應(yīng)用微纖維膠原支架構(gòu)建組織工程軟骨

【摘要】:背景:尋找理想的組織工程支架材料,仍然是軟骨組織工程學(xué)的重要課題之一。目的:探討微纖維膠原應(yīng)用于軟骨組織工程支架的可行性。方法:手術(shù)切取兔耳軟骨,提取原代軟骨細(xì)胞,并進(jìn)行擴(kuò)增培養(yǎng)。將海綿狀微纖維膠原用無(wú)菌刀片切割成小方塊,取第2代軟骨細(xì)胞混懸液接種于微纖維膠原支架材料上。體外培養(yǎng)1周后,植入裸鼠體內(nèi),觀察8周后取材,進(jìn)行大體、組織學(xué)、免疫組織化學(xué)等方法觀察。結(jié)果與結(jié)論:(1)在細(xì)胞接種過(guò)程中,濕水之后的微纖維膠原支架無(wú)明顯縮水,仍然能夠保持其立體形態(tài);(2)體外培養(yǎng)1周以及植入體內(nèi)8周后,無(wú)明顯體積和形態(tài)變化;(3)裸鼠體內(nèi)培養(yǎng)8周后形成白色半透明且富有彈性的成熟軟骨塊;(4)組織學(xué)觀察可見所構(gòu)建的軟骨組織成熟,基質(zhì)分泌旺盛,組織內(nèi)仍可見未完全降解的微纖維膠原材料;(5)結(jié)果表明,微纖維膠原蛋白是一種良好的軟骨組織工程支架材料。
[Abstract]:Background: searching for ideal scaffold materials for tissue engineering is still one of the important topics in cartilage tissue engineering. Objective: to explore the feasibility of the application of microfiber collagen in cartilage tissue engineering scaffolds. The primary chondrocytes were extracted and cultured. The spongy microfibrous collagen was cut into small cubes with aseptic blades, and the second passage chondrocytes were mixed with the scaffold of microfiber collagen. After 1 week of culture in vitro, the chondrocytes were cultured for 1 week. Results and conclusion: during cell inoculation, microfiber collagen scaffolds did not shrink significantly after wetting water, and the microfibrillar collagen scaffolds did not shrink after wetting water, and the microfibrillar collagen scaffolds were taken from nude mice after 8 weeks of observation, and were observed by gross, histological and immunohistochemical methods. Still able to maintain its stereomorphology for 1 week in vitro and 8 weeks after implantation. After 8 weeks of culture in nude mice, white translucent and elastic mature chondrocytes were formed. Histological observation showed that the chondrocytes were mature and exuberant in matrix secretion. The results showed that microfibrillar collagen was a good scaffold for cartilage tissue engineering.
