微加工技術(shù)在細(xì)胞形狀和排列控制中的應(yīng)用

【摘要】:背景:精確控制細(xì)胞形狀、實(shí)現(xiàn)細(xì)胞體外排列對(duì)細(xì)胞生物學(xué)、組織工程和再生醫(yī)學(xué)研究及應(yīng)用均有極其重要的意義,而目前對(duì)控制細(xì)胞幾何形狀和誘導(dǎo)體外排列的方法缺乏系統(tǒng)性的研究和表述。目的:總結(jié)并討論表面化學(xué)修飾技術(shù)、表面拓?fù)鋵W(xué)改造技術(shù)加工制備細(xì)胞生長(zhǎng)基底的方法,以及實(shí)現(xiàn)細(xì)胞形狀和排列控制的可行性和最新進(jìn)展。方法:作者檢索1995至2017年百鏈云數(shù)據(jù)庫(kù)和Pub Med數(shù)據(jù)庫(kù)中與微加工方法對(duì)細(xì)胞形狀和體外排列影響相關(guān)的文獻(xiàn),并進(jìn)行系統(tǒng)整理、歸納總結(jié)和分析。結(jié)果與結(jié)論:隨著微納米加工技術(shù)的發(fā)展和對(duì)細(xì)胞生物學(xué)行為的深入研究,表面化學(xué)修飾、表面拓?fù)鋵W(xué)改造、纖維支架等方法逐步被建立,以實(shí)現(xiàn)細(xì)胞形狀的精確控制和體外排列。表面微圖案化、微柱、微孔陣列等拓?fù)浣Y(jié)構(gòu)能夠精確控制細(xì)胞幾何形狀,而基底表面拓?fù)湫蚊矊?duì)細(xì)胞排列占據(jù)更重要的支配作用,可將基底表面化學(xué)修飾作用和拓?fù)湫蚊沧饔孟嘟Y(jié)合更好地實(shí)現(xiàn)細(xì)胞體外排列。合適的基底結(jié)構(gòu)可用于精確控制細(xì)胞的形狀、調(diào)控干細(xì)胞的分化抑或誘導(dǎo)細(xì)胞的定向排列、促進(jìn)細(xì)胞融合和分化等。
[Abstract]:Background: the precise control of cell shape, the arrangement of the cells in vitro cell biology, significance of tissue engineering and regenerative medicine research and application are very important, and the control of cell shape and arrangement method of in vitro induced by the lack of research and expression system. Objective: To summarize and discuss the surface chemical modification technology, processing technology of surface topology transformation the preparation method of basal cell growth, and the feasibility of cell shape and arrangement of the control and the latest progress. Methods: the authors retrieved from 1995 to 2017 100 chain cloud database and Pub Med data processing method and micro arrangement influences relevant literature on cell shape and in vitro in library, and systematically summarized and analyzed. Results and conclusion: with the in-depth study of the development of micro nano machining technology and on cell biological behavior, surface modification, surface topology transformation, Fiber support gradually is established, and arranged to achieve precise control of in vitro cell shape. The surface micro patterning, micro column, micro hole array topology can accurately control the cell geometry, topology and surface morphology of the basal cells arranged more important to occupy the dominant role, the substrate surface chemical modification and topology function with better cell arrangement. The basement structure suitable for precise control of cell shape, the regulation of stem cell differentiation or induce cell alignment, promote cell fusion and differentiation.
