SMAS修飾的PEGDA載電凝膠對MC3T3-E1細胞粘附和增殖的影響

【摘要】:目的:觀察甲基丙烯磺酸鈉(SMAS)修飾的聚乙二醇雙丙烯酸酯(PEGDA)水凝膠對小鼠前成骨細胞(MC3T3-E1細胞)粘附及增殖的影響。方法:通過在PEGDA凝膠前體溶液中加入不同濃度的SMAS小分子,合成出攜帶不同電量負電荷的新型載電凝膠。掃描電鏡觀察各組水凝膠的表面結構。檢測各組水凝膠的蛋白吸附量。觀察種子細胞在各組水凝膠表面的早期粘附,并檢測不同時間點下凝膠表面細胞粘附量。用cck8法檢測細胞在凝膠表面的增殖情況。結果:隨著SMAS濃度的提高,凝膠的蛋白吸附量不斷提高,同時隨著SMAS修飾量的提高,細胞在凝膠表面得到更好的伸展,同時凝膠表面的細胞粘附數(shù)明顯提高(P0.05)。而在細胞增殖方面,cck8結果表明隨著SMAS修飾量的提高,凝膠表面的細胞增殖也得到明顯改善。結論:SMAS小分子修飾的PEGDA凝膠能夠有效地促進MC3T3-E1細胞在其表面的粘附和增殖。
[Abstract]:Objective: to observe the effect of PEGDAA hydrogel modified with sodium methallyl sulfonate (SMASS) on mouse preosteoblast MC3T3-E1 cells. Methods: different concentrations of SMAS small molecules were added to the solution of PEGDA gel precursor. The surface structure of hydrogels in each group was observed by scanning electron microscope. The protein adsorption capacity of hydrogels in each group was measured. The early adhesion of seed cells to the surface of hydrogels in each group was observed. . The amount of cell adhesion on the gel surface was measured at different time points. The cell proliferation on the gel surface was detected by cck8 method. Results: with the increase of SMAS concentration, the amount of protein adsorbed on the gel increased. At the same time, with the increase of the amount of SMAS modification, the cells extended better on the gel surface, and the cell adhesion number on the gel surface increased significantly, while the cell proliferation. Cck8 results showed that with the increase of SMAS modification amount. Conclusion the PEGDA gel modified with small molecular weight of PEGDA can effectively promote the adhesion and proliferation of MC3T3-E1 cells on its surface.
