目的:探討微小RNA-541-3p（miR-541-3p）對皮膚黑色素瘤細胞增殖、遷移、侵襲的影響及其分子機制。方法:采用實時熒光定量聚合酶鏈反應(yīng)（qRT-PCR）與蛋白免疫印跡法（Western blotting）分別檢測皮膚黑色素瘤組織中miR-541-3p、SENP1的表達水平;以人皮膚黑色素瘤細胞A375為研究對象,分別將miR-NC、miR-541-3p mimics、si-NC、si-SENP1、miR-541-3p mimics與pcDNA、miR-541-3p mimics與pcDNA-SENP1轉(zhuǎn)染至A375細胞;甲基噻唑基四唑（MTT）檢測細胞增殖;Transwell小室實驗檢測細胞遷移及侵襲能力;雙熒光素酶報告實驗驗證miR-541-3p過表達對野生型和突變型SENP1熒光素酶活性的影響;Western blotting檢測細胞周期蛋白1（CyclinD1）、p21、基質(zhì)金屬蛋白酶-2（MMP-2）、基質(zhì)金屬蛋白酶-9（MMP-9）蛋白表達量。結(jié)果:與正常皮膚組織比較,皮膚黑色素瘤組織中miR-541-3p的表達量顯著降低（P<0.05）,SENP1 mRN...
