Psoriasin在扁平苔蘚、急性發(fā)熱性嗜中性皮病中的表達(dá)和意義

【摘要】:Psoriasin為S100蛋白家族成員,于1991年在銀屑病患者皮損中首次發(fā)現(xiàn)。20年來(lái)隨著不斷研究,psoriasin在特應(yīng)性皮炎、蕈樣肉芽腫等多種炎癥鱗屑性皮膚病及皮膚癌癥等疾病中被發(fā)現(xiàn),其不僅與角質(zhì)細(xì)胞增殖分化有關(guān),并且具有趨化中性粒細(xì)胞和CD4+T細(xì)胞至角質(zhì)層的生物學(xué)功能,近年來(lái)還發(fā)現(xiàn)psoriasin參與了皮膚的抗菌、調(diào)節(jié)免疫等作用。Psoriasin與皮膚病的相關(guān)性越來(lái)越受到重視,但是其在炎癥性皮膚病中的具體作用機(jī)制尚不明確,對(duì)于真皮層炎癥的作用尚有待進(jìn)一步研究。 目的: 探討psoriasin在扁平苔蘚、急性發(fā)熱性嗜中性皮病中的表達(dá)情況;了解psoriasin在炎癥性皮膚病中的作用;為明確psoriasin在疾病中的作用機(jī)制提供理論依據(jù)。 方法: 選取吉林大學(xué)第二醫(yī)院皮膚科近16年(1996.06-2011.03)經(jīng)臨床及皮膚組織病理學(xué)明確診斷為扁平苔蘚及急性發(fā)熱性嗜中性皮病的石蠟包埋組織標(biāo)本,應(yīng)用免疫組織化學(xué)方法,觀察psoriasin在上述疾病組織中的表達(dá)情況。 結(jié)果: Psoriasin在正常皮膚組織中未見(jiàn)表達(dá),在扁平苔蘚中及急性發(fā)熱性嗜中性皮病中均有陽(yáng)性表達(dá)。將20例正常皮膚組織作為對(duì)照組,其中陽(yáng)性0例,陰性20例。在30例扁平苔蘚中,psoriasin陽(yáng)性28例(93.33%),陰性2例(6.67%),,與對(duì)照組比較有顯著性差異(p0.01);在30例急性發(fā)熱性嗜中性皮病中,psoriasin陽(yáng)性26例(86.67%),陰性4例(13.33%),與對(duì)照組比較有顯著性差異(p0.01)。 結(jié)論: 1、首次研究了psoriasin在扁平苔蘚、急性發(fā)熱性嗜中性皮病的表達(dá)情況。 2、免疫組織化學(xué)染色結(jié)果顯示psoriasin在扁平苔蘚和急性發(fā)熱性嗜中性皮病的表皮層均有陽(yáng)性表達(dá); 3、Psoriasin可能參與了扁平苔蘚、急性發(fā)熱性嗜中性皮病的發(fā)生、發(fā)展過(guò)程; 4、扁平苔蘚皮損中CD4+T細(xì)胞主要在真皮層浸潤(rùn),急性發(fā)熱性嗜中性皮病中中性粒細(xì)胞主要在真皮層集聚,提示psoriasin不僅能參與角質(zhì)層炎癥反應(yīng),而且可能介導(dǎo)真皮層炎癥細(xì)胞的趨化。
[Abstract]:Psoriasin, a member of the S100 protein family, was first found in psoriatic lesions in 1991. In the past 20 years, psoriasin has been found in many kinds of inflammatory scaly dermatosis and skin cancer, such as atopic dermatitis, mycosis fungoides and so on. It is not only related to the proliferation and differentiation of keratinocytes, but also has the biological function of chemotactic neutrophils and CD4 T cells to the cuticular layer. In recent years, it has also been found that psoriasin is involved in the antibacterial activity of the skin. The correlation between Psoriasin and dermatosis has been paid more and more attention, but the specific mechanism of Psoriasin in inflammatory dermatosis is not clear, and the effect of Psoriasin on dermis inflammation needs to be further studied. Objective: to investigate the expression of psoriasin in lichen planus and acute febrile neutral dermatosis, to understand the role of psoriasin in inflammatory dermatosis, and to provide theoretical basis for understanding the mechanism of psoriasin in the disease. Methods: paraffin embedded tissue specimens diagnosed as lichen planus and acute febrile neutral dermatosis were selected from the Department of Dermatology, the second Hospital of Jilin University in recent 16 years (June 06, 1996.2011.03), which were diagnosed as lichen planus and acute febrile neutral dermatosis. The expression of psoriasin in the above diseases was observed by immunohistochemistry. Results: Psoriasin was not expressed in normal skin tissue, but positive in lichen planus and acute febrile neutrophil disease. 20 cases of normal skin tissue were used as control group, of which 0 cases were positive and 20 cases were negative. In 30 cases of lichen planus, psoriasin was positive in 28 cases (93.33%) and negative in 2 cases (6.67%), which was significantly different from that in the control group (p0.01). In 30 cases of acute febrile neutral dermatosis, psoriasin was positive in 26 cases (86.67%) and negative in 4 cases (13.33%), which was significantly different from that in the control group (p0.01). Conclusion: 1. The expression of psoriasin in lichen planus and acute febrile neutrophil disease was studied for the first time. 2. 2. The results of Immunohistochemical staining showed that psoriasin was positive in the epidermis of lichen planus and acute febrile neutrophil disease. (3) Psoriasin may be involved in the occurrence and development of lichen planus and acute febrile neutrophil disease, and the expression of Psoriasin may be involved in the occurrence and development of lichen planus and acute febrile neutrophil disease. 4. CD4 T cells were mainly infiltrated in the dermis of lichen planus, and neutrophils of acute febrile neutrophils were mainly concentrated in the dermis, suggesting that psoriasin could not only participate in cuticular inflammation. It may also mediated the chemotaxis of inflammatory cells in dermis.
