【摘要】：目的:研究微管不穩(wěn)定蛋白stathmin在炎癥性皮膚病引起的慢性瘙癢患者發(fā)病機(jī)制中的作用。方法:收集35例炎癥性皮膚病引起的慢性瘙癢患者的皮損標(biāo)本和3例正常人的皮膚標(biāo)本。用免疫組化(immunohistochemistry,IHC)法檢測皮膚活檢標(biāo)本中stathmin的表達(dá)。用蘇木精-伊紅染色(hematoxylin-eosinstaining,HE染色)檢測患者表皮組織中病理結(jié)構(gòu)的變化。結(jié)果:IHC法檢測結(jié)果顯示,與正常對照的皮膚相比,stathmin在患者皮損中表皮角質(zhì)形成細(xì)胞、毛囊上皮細(xì)胞和免疫炎癥細(xì)胞中高表達(dá)。HE染色顯示慢性瘙癢患者皮損處增厚、角化過度,并伴有炎癥性細(xì)胞浸潤。結(jié)論:研究結(jié)果初步表明stathmin在慢性瘙癢的機(jī)制中發(fā)揮作用。Stathmin通過在角質(zhì)形成細(xì)胞、上皮細(xì)胞和免疫炎癥細(xì)胞中的表達(dá)影響皮膚屏障功能,促進(jìn)慢性瘙癢的進(jìn)程。Stathmin可能是慢性瘙癢治療的新靶點(diǎn),對慢性瘙癢的治療和新藥研發(fā)具有重要意義。
[Abstract]:Aim: to study the role of microtubule unstable protein (stathmin) in the pathogenesis of chronic pruritus induced by inflammatory dermatosis. Methods: the skin lesions of 35 patients with chronic pruritus caused by inflammatory dermatosis and 3 normal skin specimens were collected. Immunohistochemical (immunohistochemistry,IHC) method was used to detect the expression of stathmin in skin biopsy specimens. Hematoxylin-eosin staining (hematoxylin-eosinstaining,HE) was used to detect the pathological changes in the epidermis of the patients. Results: the results of IHC assay showed that stathmin was highly expressed in keratinocytes, hair follicle epithelial cells and immune inflammatory cells in the lesions of patients with chronic pruritus compared with the normal skin. He staining showed that the lesions of patients with chronic pruritus were thickened. Hyperkeratosis is associated with inflammatory cell infiltration. Conclusion: the results suggest that stathmin plays a role in the mechanism of chronic pruritus. Stathmin affects skin barrier function through the expression of stathmin in keratinocytes, epithelial cells and immune inflammatory cells. Stathmin may be a new target for the treatment of chronic pruritus, which is of great significance to the treatment of chronic pruritus and the development of new drugs.
