【摘要】：目的觀察內(nèi)源性omega-3多元不飽和脂肪酸(polyunsaturated fatty acids,PUFAs)對小鼠銀屑病樣皮損以及脾臟Th17細(xì)胞因子的影響。方法本實(shí)驗(yàn)采用Fat-1轉(zhuǎn)基因小鼠模型,將野生型(Wild-type)小鼠和Fat-1小鼠各分為2組,背部以及右耳背外涂咪喹莫特乳膏(Imiquimod,IMQ)和凡士林乳膏(cream),建立銀屑病樣皮損模型及對照組。觀察4組小鼠給藥部位皮膚變化,對表皮組織進(jìn)行病理切片和HE染色,流式細(xì)胞術(shù)測定脾細(xì)胞內(nèi)Th17細(xì)胞因子的百分比。結(jié)果 Fat-1 IMQ與WT IMQ組相比,表皮因IMQ作用而出現(xiàn)角質(zhì)層、棘細(xì)胞層的增厚,大體表現(xiàn)的紅斑、增厚、鱗屑反應(yīng)均較后者輕微,同時(shí)IMQ所造成的脾細(xì)胞內(nèi)Th17細(xì)胞以及炎癥因子IL-17升高,也在Fat-1組小鼠中呈現(xiàn)出了較低的水平(P0.05)。而兩組cream組間差異無統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論內(nèi)源性omega-3 PUFAs在銀屑病中能夠減少脾細(xì)胞內(nèi)Th17細(xì)胞的水平,減少IL-17炎癥因子的分泌以及改善銀屑病樣紅斑、鱗屑、增厚等臨床癥狀,從而發(fā)揮治療銀屑病的作用。
[Abstract]:Objective to observe the effects of endogenous omega-3 polyunsaturated fatty acid (polyunsaturated fatty acids,PUFAs) on psoriatic lesions and Th17 cytokines in spleen of mice. Methods Fat-1 transgenic mice were used in this experiment. Wild type (Wild-type) mice and Fat-1 mice were divided into two groups: the back and the right ear were coated with Imiquimod Cream (Imiquimod,IMQ) and Vaseline Cream (cream), respectively. To establish psoriasis-like skin lesion model and control group. The skin changes of the four groups were observed, the epidermal tissues were stained with HE and pathological sections, and the percentage of Th17 cytokines in spleen cells was determined by flow cytometry. Results compared with the WT IMQ group, the keratinocytes appeared in the epidermis due to the action of IMQ, the thickening of the spinous cell layer, the erythema, thickening and the scale reaction of the Fat-1 IMQ group were more slight than those of the latter group. At the same time, the increase of Th17 cells and inflammatory factor IL-17 in spleen cells caused by IMQ also showed a lower level in Fat-1 group (P0.05). There was no significant difference in cream between the two groups (P0.05). Conclusion Endogenous omega-3 PUFAs can reduce the level of Th17 cells in splenic cells, decrease the secretion of IL-17 inflammatory factors and improve the clinical symptoms of psoriasis like erythema, scales and thickening in psoriasis, so as to play a role in the treatment of psoriasis.
