【摘要】： 目的:檢測人類皰疹病毒8型(HHV-8)在四種非Kaposi肉瘤皮損組織中的感染情況,以初步探討HHV-8與皮膚鱗狀細(xì)胞癌、蕈樣肉芽腫、Castleman病和化膿性肉芽腫相關(guān)性研究。方法:使用酚-氯仿-異戊醇方法對新疆維吾爾自治區(qū)人民醫(yī)院皮膚性病科、病理科從2004年到2009年收集的30例鱗狀細(xì)胞癌、20例Castleman病(其中5例為多中性Castleman病)、13例蕈樣肉芽腫、20例化膿性肉芽腫石蠟包埋組織進(jìn)行HHV-8 DNA抽提;然后使用巢式PCR方法對抽提的DNA進(jìn)行ORF26基因片段的擴(kuò)增,擴(kuò)增產(chǎn)物在1.5%的瓊脂糖凝膠上電泳,并在凝膠成像系統(tǒng)下觀察電泳結(jié)果,從而確定ORF26基因陽性標(biāo)本;然后計算HHV-8在各種標(biāo)本中的陽性率。結(jié)果:30例皮膚鱗狀細(xì)胞癌皮損組織有5份為陽性(16.7%)。13例蕈樣肉芽腫石蠟組織有1例為陽性(7.7%),20例化膿性肉芽腫和20例Castleman病石蠟組織中均未檢出HHV-8。結(jié)論:①HHV-8可能是新疆皮膚鱗狀細(xì)胞癌發(fā)病原因之一。②HHV-8與蕈樣肉芽腫的發(fā)病可能不具有相關(guān)性,HHV-8可能是MF發(fā)生后偶發(fā)的一種附帶感染。③Castleman病和化膿性肉芽腫的發(fā)病可能均與HHV-8無關(guān)。
[Abstract]:Objective: to detect the infection of human herpesvirus 8 (HHV-8) in four kinds of non Kaposi sarcomas and to explore the relationship between HHV-8 and cutaneous squamous cell carcinoma, mycosis fungoides, Castleman disease and suppurative granuloma. Methods: using phenol-chloroform-isoamyl alcohol method, 30 cases of squamous cell carcinoma (SCC) and 20 cases of Castleman disease (5 cases of polyneutral Castleman disease) were collected from 2004 to 2009 in the Department of Dermatology and venereal Diseases of the people's Hospital of Xinjiang Uygur Autonomous region. 13 cases of fungoid granuloma and 20 cases of suppurative granuloma paraffin embedded tissue were extracted by HHV-8 DNA. Then nested PCR method was used to amplify the ORF26 gene fragment of extracted DNA. The amplified product was electrophoresis on 1.5% agarose gel. The results of electrophoresis were observed under the gel imaging system to determine the positive samples of ORF26 gene. Then the positive rate of HHV-8 in various specimens was calculated. Results: 5 cases (16.7%) were positive in skin lesions of 30 cases of cutaneous squamous cell carcinoma and 1 case (7.7%) of paraffin tissues of 13 cases of mycosis fungoides. HHV-8. was not detected in 20 cases of suppurative granuloma and 20 cases of Castleman's disease in paraffin tissues. Conclusion: 1HHV-8 may be one of the causes of cutaneous squamous cell carcinoma in Xinjiang. 2HHV-8 may not be associated with the pathogenesis of mycosis fungoides. HHV-8 may be an incidental infection after MF. 3Castleman disease and purulent granuloma may not be associated with HHV-8.
