【摘要】：目的探討Neuropilin-1在嬰幼兒皮膚血管瘤增生期、消退期組織中的表達及意義。 材料與方法收集華中科技大學附屬協(xié)和醫(yī)院2007年1月—2010年3月間切取的47例患者的血管瘤組織作為實驗組,術(shù)前均未行其他治療并經(jīng)病理檢查和追溯病史證實為嬰幼兒血管瘤。對照組為我科在手術(shù)時所留取的含血管的正常皮膚組織,共10例。根據(jù)HE染色結(jié)果將血管瘤分為增生期組與消退期組,采用免疫組化SABC技術(shù),檢測26例增生期嬰幼兒血管瘤、21例消退期及10例正常皮膚組織中Neuropilin-1蛋白的表達,應用Image-pro plus6.0軟件測量各標本Neuropilin-1蛋白表達的積分光密度(IOD)、平均光密度,結(jié)果用SPSS12.0統(tǒng)計軟件進行數(shù)據(jù)分析。 結(jié)果47例嬰幼兒血管瘤標本中增生期組、消退期組Neuropilin-1均呈陽性表達,正常皮膚血管內(nèi)皮細胞未見明顯表達。增生期血管瘤中Neuropilin-1的表達高于消退期和正常皮膚,其差異有顯著性(P0.05);消退期表達水平高于正常皮膚組織,其差異有顯著性(P0.05)。 結(jié)論Neuropilin-1在嬰幼兒血管瘤中高表達,增生期與消退期表達水平存在顯著差異,可能是血管瘤內(nèi)皮細胞增殖的重要促進因子。
[Abstract]:Objective to investigate the expression and significance of Neuropilin-1 in the proliferative and involutive phases of infantile skin hemangioma. Materials and methods the hemangioma tissues of 47 patients were collected from Union Hospital of Huazhong University of Science and Technology from January 2007 to March 2010 as experimental group. No other treatment was performed before operation and proved to be infantile hemangioma by pathological examination and retrospective history. The control group consisted of 10 cases of normal skin tissue containing blood vessels. According to the results of HE staining, hemangiomas were divided into proliferative phase group and regression phase group. The expression of Neuropilin-1 protein in 26 cases of proliferative phase hemangioma, 21 cases of regression phase and 10 cases of normal skin tissue were detected by immunohistochemical SABC technique. The integral optical density (IOD),) of Neuropilin-1 protein was measured by Image-pro plus6.0 software. The results were analyzed by SPSS12.0 software. Results in 47 cases of infantile hemangioma, the positive expression of Neuropilin-1 was found in proliferative phase group and regression phase group, but no obvious expression was found in normal skin vascular endothelial cells. The expression of Neuropilin-1 in proliferative hemangioma was higher than that in involutive phase and normal skin (P0.05), and the expression level in involutive phase was higher than that in normal skin tissue (P0.05). Conclusion the high expression of Neuropilin-1 in infantile hemangioma and the significant difference between proliferative phase and regression phase may play an important role in the proliferation of endothelial cells in hemangioma.
