阿維A酸對小鼠黑色素瘤細胞株B16增殖和Livin、bFGF表達的影響

【摘要】: 目的:惡性黑色素瘤(malignant malanoma,MM)是一種主要發(fā)生于皮膚由黑色素細胞惡化形成的能夠產(chǎn)生黑色素嚴(yán)重威脅人們生命的高度惡性腫瘤,其生物學(xué)行為高度惡性,具有侵襲組織和持續(xù)增殖能力,并可逃避免疫監(jiān)視。目前為止還沒有有效的預(yù)防治療方法,外科手術(shù)仍然是現(xiàn)在治療黑色素瘤的首選方法,但是即便外科手術(shù)切除,預(yù)后仍很不理想,進展期此腫瘤患者平均的生存時間僅為6~9個月。因此,初步研究阿維A酸治療惡性黑色素瘤的作用及其機制,探討作用機制是否與阿維A酸影響Livin、bFGF的表達有關(guān),進而為臨床誘導(dǎo)分化治療黑色素瘤提供實驗基礎(chǔ)。 維甲酸類(retinoids)包括維生素A的天然和人工合成的衍生物。它對上皮細胞有抑制其異常增殖和分化的作用,并可促進上皮組織正常角化,溶解角質(zhì)使其恢復(fù)正常的作用,在臨床上已用來預(yù)防及治療多種皮膚病。近來科學(xué)研究表明,維甲酸還具有很強的誘導(dǎo)分化抑制腫瘤細胞增殖的作用,具有明顯的時間依賴性和濃度依賴性。 方法: 1細胞培養(yǎng) 在含10%胎牛血清的RPMI-1640培養(yǎng)基(含青霉素100U/ml,含鏈霉素100U/ml,PH7.2)中培養(yǎng)B16細胞,置于飽和濕度、37℃、5%CO2的培養(yǎng)箱內(nèi)做常規(guī)傳代培養(yǎng)。 2四甲基偶氮唑藍(MTT)比色法 檢測阿維A對B16細胞增殖的抑制作用。采用濃度不同的阿維A(0.1 umol/L、1umol/L、10umol/L、100umol/L)作用不同時間(24、48、72小時)后,檢測對B16細胞增殖的抑制作用。 3倒置相差顯微鏡及HE染色法 觀察經(jīng)過不同濃度藥物處理后B16細胞的分化及形態(tài)變化并同時把細胞HE染色后進行觀察。 4免疫細胞化學(xué)s-p法 檢測經(jīng)不同濃度藥物處理后的B16細胞Livin及堿性成纖維細胞生長因子(bFGF)蛋白的表達狀況。 5流式細胞分析技術(shù) 檢測細胞周期變化及凋亡的進展,將實驗分為5組,分別為陰性對照組(不加任何藥物),阿維A濃度分別為0.1 umol/L,1 umol/L, 10umol/L,100 umol/L各一組。各組不同濃度藥物處理48小時之后,收集細胞,應(yīng)用流式細胞儀,檢測各組細胞周期分布、細胞凋亡情況及Livin、bFGF蛋白表達狀況。 6統(tǒng)計軟件SPSS13.0對數(shù)據(jù)進行統(tǒng)計分析。 結(jié)果: 1 MTT比色分析法檢測 結(jié)果顯示阿維A酸對B16細胞增殖有顯著的抑制作用(相對抑制率),各個組間的差別有顯著性,并且不同濃度阿維A酸作用于小鼠B16細胞后所產(chǎn)生的抑制作用表現(xiàn)出顯著的時間依賴效應(yīng)關(guān)系和劑量依賴效應(yīng)關(guān)系。 2阿維A酸對B16細胞形態(tài)及分化的影響 陰性對照組:B16細胞呈梭形或多角形,貼壁生長良好,透明,折光性好,增殖旺盛;經(jīng)阿維A酸處理B16細胞后,細胞變圓,貼壁差,生長不佳,折光性減弱,核固縮現(xiàn)象增多,皺縮,并且出現(xiàn)小部分無細胞生長區(qū)及細胞漂浮等現(xiàn)象增多,而且隨著阿維A處理時間的延長及濃度的增加上述現(xiàn)象變得更加明顯嚴(yán)重。 3免疫細胞化學(xué)s-p法檢測B16細胞Livin、bFGF蛋白表達情況 Livin、bFGF蛋白主要表達于細胞漿內(nèi),DAB染色為深棕黃色。經(jīng)不同濃度藥物處理后的細胞蛋白陽性表達的程度不同,陰性對照組細胞表達較強,隨著藥物濃度的增加染色程度都有不同程度的減弱,并且差異有顯著性(p0.05)。 4流式細胞術(shù)檢測經(jīng)不同濃度藥物作用后,細胞的周期、凋亡變化 流式細胞術(shù)檢測顯示:G1期延滯,G1-S期轉(zhuǎn)化抑制,G0/G1期的比例上升,S期細胞的比例減少。 5流式細胞術(shù)測定不同濃度藥物作用后,B16細胞Livin、bFGF蛋白的表達 經(jīng)不同濃度藥作用作用后,與陰性對照組相比,表達Livin、bFGF的陽性細胞數(shù)逐漸降低,經(jīng)阿維A酸作用細胞48小時后,各組Livin蛋白表達熒光指數(shù)(FI)分別為1.6500、1.4215、1.2100、1.1124,bFGF蛋白表達熒光指數(shù)(FI)分別為1.8421、1.6446、1.4300、1.2218。把數(shù)據(jù)分別進行兩兩比較,之間的差異均有顯著性(p0.05)。 結(jié)論: 1維A酸可以抑制B16細胞增殖,而且在一定時間和濃度范圍內(nèi),對B16細胞增殖的抑制具有時間依賴性和量效依賴性。 2四個濃度的阿維A酸均可以影響小鼠黑色素瘤細胞B16從G0/G1期進入S期,然而誘導(dǎo)B16細胞凋亡的作用并不十分得明顯。 3阿維A酸對B16細胞增殖的抑制作用可能與下調(diào)凋亡抑制因子Livin和堿性成纖維因子bFGF的表達有關(guān)。這有可能是其抗腫瘤增殖和誘導(dǎo)癌細胞分化的機制之一。
