【摘要】：目的研究AT-序列結(jié)合蛋白1(STAB1)基因?qū)θ似つw癌細(xì)胞A431凋亡及侵襲能力的影響。方法以人皮膚癌細(xì)胞A431為研究對(duì)象,根據(jù)不同處理方法分為空白組(A431組)、對(duì)照組(NC-siRNA組)和實(shí)驗(yàn)組(STAB1-siRNA組)。用lipofectamine 2000脂質(zhì)體分別轉(zhuǎn)染無(wú)關(guān)小RNA(NC-siRNA)和針對(duì)STAB1的小干擾RNA(STAB1-siRNA)后,計(jì)數(shù)A431細(xì)胞、NC-siRNA A431和STAB1-siRNA A431細(xì)胞的增殖情況、細(xì)胞凋亡比例和3組細(xì)胞的遷移能力。結(jié)果從48 h開始,小干擾RNA下調(diào)STAB1基因后,A431細(xì)胞增殖能力顯著降低(P0.05);STAB1-siRNA細(xì)胞凋亡比例明顯增高(P0.05);A431、NC-siRNA A431及STAB1-siRNA A431細(xì)胞劃痕后0,48 h在下調(diào)STAB1基因后,皮膚癌細(xì)胞遷移能力顯著降低(P0.05)。結(jié)論小干擾RNA下調(diào)STAB1基因表達(dá)后可促進(jìn)人皮膚癌細(xì)胞A431凋亡,抑制細(xì)胞增殖和遷移能力。
[Abstract]:Objective to study the effect of AT- sequence binding protein 1 (STAB1) gene on apoptosis and invasion of human skin cancer cell A431. Methods Human skin cancer cell A431 was divided into blank group (A431 group), control group (NC-siRNA group) and experimental group (STAB1-siRNA group) according to different treatment methods. After transfection of small RNA (NC-siRNA) and small interfering RNA (STAB1-siRNA) targeting STAB1 with lipofectamine 2000 liposome, the proliferation of A431 cells, NC-siRNA A431 and STAB1-siRNA A431 cells, the percentage of apoptosis and the migration ability of the three groups were counted. Results after down-regulation of STAB1 gene by small interfering RNA for 48 h, the proliferation ability of A431 cells decreased significantly (P0.05), the percentage of apoptosis of STAB1-siRNA cells increased significantly (P0.05), and the migration ability of skin cancer cells decreased significantly (P0.05) at 048 h after STAB1 gene was down-regulated by A431 NC-siRNA A431 and STAB1-siRNA A431 cells. Conclusion small interfering RNA can promote the apoptosis of human skin cancer cell line A431 and inhibit cell proliferation and migration after down-regulation of STAB1 gene expression.
