ALA聯(lián)合IPL的光動力療法對豚鼠光老化模型治療作用的研究

【摘要】: 目的探討5-氨基酮戊酸(ALA)聯(lián)合強(qiáng)脈沖光(IPL)的光動力學(xué)療法(PDT)對豚鼠光老化模型的影響,并摸索合適的ALA治療濃度。 方法通過模擬日光光源(SSR)照射建立豚鼠光老化模型,照射后分別外用10%或20%ALA聯(lián)合IPL治療,每3周一次共3次。以蘇木素—伊紅染色、Weigert染色、Siriured染色觀察真皮細(xì)胞外基質(zhì)(膠原纖維、彈力纖維)的改變,以透射電鏡觀察真皮超微結(jié)構(gòu)(成纖維細(xì)胞、彈力纖維和膠原纖維)的改變,并與未接受SSR照射的同齡陰性對照組及SSR照射后同齡自然恢復(fù)組比較。 結(jié)果豚鼠在以松香蜂蠟混合液脫毛后接受亞紅斑量SSR照射,每周三次,連續(xù)照射13周,光照部位真皮淺層出現(xiàn)了典型的“光化性彈力纖維變性”的損害;彈力纖維排列紊亂、斷裂及異常彈力纖維樣物質(zhì)沉積;Ⅰ、Ⅲ型膠原纖維斷裂,排列紊亂;成纖維細(xì)胞線粒體空泡化,粗面內(nèi)質(zhì)網(wǎng)稀疏。10%ALA聯(lián)合IPL治療三次后可顯示改善上述破壞,真皮淺層嗜堿性物質(zhì)堆積減少,Ⅰ、Ⅲ型膠原纖維和彈力纖維排列趨于整齊,并且多是長纖維,斷裂減少。成纖維細(xì)胞線粒體空泡化減輕并伴有內(nèi)質(zhì)網(wǎng)擴(kuò)張,其療效顯著優(yōu)于基質(zhì)霜聯(lián)合IPL治療部位。20%ALA聯(lián)合IPL治療雖然也有促進(jìn)上述損傷后修復(fù)的作用,但并沒有進(jìn)一步提高療效,反而使部分區(qū)域“光化性彈力纖維變性”的損害進(jìn)一步加重,成纖維細(xì)胞線粒體空泡化改變普遍且明顯。 結(jié)論ALA聯(lián)合IPL的光動力療法對光老化有一定改善作用,其療效優(yōu)于僅用IPL治療。選擇合適的ALA治療濃度非常關(guān)鍵,豚鼠的動物實驗提示10%ALA可能為較合適的濃度。
[Abstract]:Objective to investigate the effect of photodynamic therapy of 5-aminolevulinic acid (ALA) combined with intense pulse light (IPL) on photoaging of guinea pigs, and to explore the appropriate concentration of ALA. Methods the photoaging model of guinea pigs was established by simulated solar light source (SSR) irradiation. After irradiation, 10% or 20%ALA combined with IPL were treated externally for 3 times every 3 weeks. The changes of extracellular matrix (collagen fiber, elastic fiber) and ultrastructure (fibroblast, elastic fiber and collagen fiber) of dermis were observed by Weigert staining with hematoxylin and eosin staining. The results were compared with those of the same age negative control group without SSR irradiation and the natural recovery group of the same age after SSR irradiation. Results Guinea pigs were exposed to suberythema SSR after depilation with rosin wax mixture, three times a week for 13 weeks. The typical "photoelastic fiber denaturation" was observed in the superficial dermis of the illumination area, and the arrangement of elastic fibers was disordered.
