負(fù)載核糖體蛋白L8的樹突狀細(xì)胞對黑色素瘤的抑制作用

【摘要】:背景與目的:研究發(fā)現(xiàn)核糖體蛋白L8(ribosomal protein L8,RPL8)在黑色素瘤中表達(dá)能激活黑色素瘤患者外周血單個核細(xì)胞增殖并產(chǎn)生白細(xì)胞介素2,提示RPL8可能參與抗腫瘤免疫應(yīng)答,有望成為抗腫瘤治療的靶點。本研究通過RPL8蛋白負(fù)載樹突狀細(xì)胞(dentritic cell,DC),探討負(fù)載RPL8蛋白的DC對黑色素瘤的免疫效應(yīng)。方法:原核表達(dá)RPL8蛋白,純化后致敏小鼠骨髓來源DC,流式細(xì)胞儀檢測DC表面標(biāo)志,MTT法檢測細(xì)胞毒性T淋巴細(xì)胞對小鼠黑色素瘤細(xì)胞的殺傷作用;負(fù)載RPL8蛋白DC免疫治療小鼠后,觀察腫瘤體積變化及小鼠生存時間。結(jié)果:純化蛋白經(jīng)蛋白質(zhì)印跡法(Western blot)分析見約28×103大小的特異性條帶;DC經(jīng)RPL8及細(xì)菌脂多糖(Lipoplysaccharide,LPS)誘導(dǎo)成熟后細(xì)胞表面CD11c、CD80、MHC-Ⅰ類、MHC-Ⅱ類分子表達(dá)增高,能激活T淋巴細(xì)胞,對B16細(xì)胞有抑制作用,RPL8-DC組的抑制率在效靶比為30∶1時高達(dá)70%,較PBS組和DC組明顯增高;負(fù)載RPL8蛋白DC免疫治療小鼠后,腫瘤體積縮小,小鼠的生存期明顯延長。結(jié)論:負(fù)載RPL8的DC對黑色素瘤有生長抑制作用。
[Abstract]:Background & AIM: the expression of ribosomal protein L8 (ribosomal protein L8 in melanoma can activate the proliferation of peripheral blood mononuclear cells and produce interleukin 2 in melanoma patients, suggesting that RPL8 may be involved in the anti-tumor immune response. It is expected to be the target of anti-tumor therapy. In this study, RPL8 protein loaded dendritic cells (dentritic cells) were used to investigate the immunological effect of RPL8 loaded DC on melanoma. Methods: RPL8 protein was expressed in prokaryotic cells, and DCderived from bone marrow of sensitized mice was purified. MTT assay was used to detect the cytotoxic T lymphocyte cytotoxicity to melanoma cells, and the DC loaded with RPL8 protein was immunized to mice. Tumor volume and survival time of mice were observed. Results: the purified protein was analyzed by Western blotting.
