IL-36α對銀屑病小鼠皮膚病變的影響及其作用機制

【摘要】:目的探討白細胞介素36α(IL-36α)對銀屑病小鼠皮膚病變的影響及其作用機制。方法將30只雌性小鼠隨機分為對照組、模型組及觀察組,每組10只。模型組及觀察組于背部裸露皮膚處涂抹5%咪喹莫特乳膏62.5 mg,對照組涂抹等量凡士林乳膏,1次/d,連續(xù)8天。觀察組隔日皮下注射IL-36α5μg,對照組、模型組注射等量PBS,共注射4次。評估各組建模第1~8天銀屑病皮損面積和嚴重程度評分(PASI評分)。建模第8天處死,測量表皮垂直厚度,分別采用實時熒光定量PCR法、Western blotting法檢測皮膚組織血管內(nèi)皮生長因子(VEGF)和IL-36αmRNA和蛋白相對表達量。結(jié)果隨建模時間延長,模型組和觀察組PASI評分逐漸升高(P均0.05)。觀察組、模型組建模第5天開始PASI評分明顯升高,以觀察組升高更明顯(P均0.05)。觀察組表皮垂直厚度為(98.519±3.442)μm,模型組為(68.383±2.499)μm,對照組為(9.804±1.682)μm,組間兩兩比較P均0.05。觀察組、模型組和對照組皮膚組織VEGF、IL-36αmRNA和蛋白相對表達量依次降低,組間兩兩比較P均0.05。結(jié)論IL-36α可加重銀屑病小鼠皮膚病變,其機制可能與促進VEGF表達有關(guān)。
[Abstract]:Objective to investigate the effect and mechanism of interleukin 36 偽 (IL 36 偽) on psoriasis in mice. Methods 30 female mice were randomly divided into control group, model group and observation group with 10 mice in each group. The model group and the observation group were smeared with 5% Imiquimod cream 62.5 mg on the bare skin of the back, while the control group was smeared with the same amount of vaseline cream once a day for 8 days. IL-36 偽 5 渭 g was injected subcutaneously every other day in the observation group, and PBS4 times in the control group and the model group. The psoriasis lesion area and severity score (PASI score) were evaluated on the 1st day of modeling in each group. On the 8th day of modeling, the vertical thickness of epidermis was measured, and the relative expressions of vascular endothelial growth factor (VEGF) and IL-36 偽 mRNA and protein in skin tissue were detected by real-time fluorescence quantitative PCR and Western blotting. Results the PASI score of model group and observation group increased gradually with the prolongation of modeling time (P 0.05). In the observation group, the PASI score of the model group was significantly higher than that of the observation group (P 0.05). The vertical thickness of epidermis was (98.519 鹵3.442) 渭 m in the observation group, (68.383 鹵2.499) 渭 m in the model group and (9.804 鹵1.682) 渭 m in the control group. The relative expression of VEGF IL-36 偽 mRNA and protein in the skin tissues of the observation group, the model group and the control group decreased in turn, with a significant difference between the two groups (P 0.05). Conclusion IL-36 偽 can aggravate skin lesions in psoriatic mice, and its mechanism may be related to the promotion of VEGF expression.
