【摘要】：目的分析smad與病理性瘢痕上皮和瘢痕癌的相關(guān)性。方法采用免疫組織化學(xué)(SP法)及實(shí)時(shí)熒光定量聚合酶鏈反應(yīng)分別對(duì)病理性瘢痕組織標(biāo)本15份、瘢痕癌組織標(biāo)本15份,正常皮膚組織標(biāo)本20份進(jìn)行研究,利用圖像分析軟件,計(jì)算三組標(biāo)本平均觀密度、陽(yáng)性面積以及2-ΔΔCT數(shù)值的表達(dá)。結(jié)果 (1)smad的蛋白及mRNA在正常皮膚組織標(biāo)本及病理性瘢痕組織標(biāo)本中呈現(xiàn)陰性及弱陽(yáng)性,在瘢痕癌組織標(biāo)本呈現(xiàn)強(qiáng)陽(yáng)性表達(dá)。(2)瘢痕癌組織標(biāo)本表達(dá)水平(陽(yáng)性面積)、表達(dá)強(qiáng)度差異有統(tǒng)計(jì)學(xué)意義(P0.05),但是正常皮膚組織標(biāo)本及病理性瘢痕組織標(biāo)本之間不存在差異(P0.05)。(3)smad的蛋白及mRNA表達(dá)呈正相關(guān)關(guān)系(r=9.34 P0.05)。結(jié)論 smad與瘢痕癌的表達(dá)有關(guān)聯(lián)性,可作為瘢痕癌靶向治療的研究重點(diǎn)。
[Abstract]:Objective to analyze the correlation between smad and pathological scar epithelium and scar carcinoma. Methods Fifteen specimens of pathological scar tissue, 15 specimens of scar carcinoma and 20 specimens of normal skin were studied by immunohistochemistry and real-time fluorescence quantitative polymerase chain reaction. Image analysis software was used. The mean apparent density, positive area and the expression of 2-螖 CT were calculated. Results the protein and mRNA of SMA were negative and weakly positive in normal skin tissue and pathological scar tissue. The expression level of scar carcinoma tissue (positive area, expression intensity difference is statistically significant (P 0.05), but there is no difference between normal skin tissue and pathological scar tissue specimen.) the expression level of scar cancer tissue is strongly positive. 2) the expression level of scar carcinoma tissue is significantly different from that of normal skin tissue and pathological scar tissue (P 0.05). There was a positive correlation between the protein expression and mRNA expression of P0.05, P0.05, P0.05, and the expression of protein and mRNA was positively correlated with that of 9.34 P0.05. Conclusion smad is associated with the expression of cicatricial carcinoma.
