銀屑病患者骨髓間充質(zhì)干細胞細胞因子分泌水平及受體表達的研究

【摘要】: [目的]研究銀屑病患者骨髓間充質(zhì)干細胞(Mesenchymal stem cells, MSCs)細胞因子分泌水平及骨髓單個核細胞(Bone marrow mononuclear cells,BMMNCs)細胞因子受體的表達狀況,并與銀屑病病變程度進行相關性分析,探討銀屑病患者骨髓造血微環(huán)境的改變。 [方法]①采用密度梯度離心法分離銀屑病患者與對照組骨髓單個核細胞,通過貼壁培養(yǎng)法分離培養(yǎng)骨髓間充質(zhì)干細胞,用流式細胞儀鑒定細胞表面標志。②采用ELISA法測定骨髓MSCs培養(yǎng)上清液中白細胞介素-8(IL-8)、白細胞介素-3(IL-3)、粒細胞-巨噬細胞集落刺激因子(GM-CSF)及肝細胞生長因子(HGF)的分泌水平。③用流式細胞儀測定骨髓單個核細胞表面白細胞介素-3受體α(IL-3Rα)和肝細胞生長因子受體(HGFR)表達水平。④根據(jù)銀屑病皮損面積和嚴重程度的PASI評分法,采用pearson相關分析評估細胞因子分泌水平與銀屑病病變程度的相關性。 [結(jié)果]①流式測定顯示培養(yǎng)的骨髓間充質(zhì)干細胞高表達CD44、CD71,而HLA-DR及造血細胞表面標志CD45、CD34陰性表達,說明所培養(yǎng)的骨髓MSCs純度較高;②銀屑病患者組骨髓間充質(zhì)干細胞IL-8、IL-3、HGF分泌水平均顯著低于對照組(P0.05),兩組間GM-CSF的分泌水平無統(tǒng)計學差異(P0.05);③銀屑病患者組單個核細胞表面IL-3Rα、HGFR表達水平顯著低于對照組(P0.05);④銀屑病患者組細胞因子分泌水平與PASI評分無相關性。 [結(jié)論]①銀屑病患者骨髓造血微環(huán)境存在異常,對銀屑病的發(fā)病可能起到重要作用。②銀屑病患者骨髓MSCs細胞因子分泌水平與病變程度無相關性。
[Abstract]:[objective] to study the levels of cytokines secreted by mesenchymal stem cells, MSCs) and the expression of cytokine receptors in bone marrow mononuclear cells of psoriatic patients, and to analyze their correlation with the degree of psoriasis. To investigate the changes of bone marrow hematopoietic microenvironment in patients with psoriasis. [methods] 1Bone marrow mononuclear cells were isolated from psoriatic patients and control group by density gradient centrifugation. Bone marrow mesenchymal stem cells were isolated and cultured by adherent culture. Identification of Cell Surface Marker .2 the secretion of Interleukin-8 (IL-8), Interleukin-3 (IL-3), granulocyte-macrophage Colony stimulating Factor (GM-CSF) and Hepatocyte growth Factor (HGF) in the supernatant of Bone Marrow MSCs Culture by flow Cytometer Level .3 the expression of interleukin-3 receptor 偽 (IL-3R 偽) and hepatocyte growth factor receptor (HGFR) on the surface of bone marrow mononuclear cells was measured by flow cytometry. 4. The PASI scoring method was used according to the area and severity of psoriatic lesions. The correlation between cytokine secretion and psoriasis was evaluated by pearson correlation analysis. [results] 1 flow cytometry showed that CD44-CD71 was highly expressed in cultured bone marrow mesenchymal stem cells, while HLA-DR and hematopoietic cell surface marker CD45 and CD34 were negative, indicating that the purity of cultured bone marrow MSCs was high. 2the secretion level of IL-8 and IL-3HGF of bone marrow mesenchymal stem cells in psoriatic patients was significantly lower than that of control group (P 0.05). There was no significant difference in the secretion level of GM-CSF between the two groups. The expression of IL-3R 偽 HGFR on the surface of mononuclear cells in psoriatic patients was significantly lower than that in control group. There was no correlation between cytokine secretion and PASI score in psoriatic patients. [conclusion] 1 abnormal hematopoietic microenvironment in bone marrow of psoriatic patients may play an important role in the pathogenesis of psoriasis. 2 there is no correlation between the level of MSCs cytokine secretion and the degree of lesion in psoriatic patients.
