KG軟膏治療急性濕疹的藥效作用及部分機理研究

【摘要】:目的:觀察KG軟膏對二甲苯致小鼠耳廓急性炎癥和2,4-二硝基氟苯(DNFB)所致遲發(fā)型超敏反應(yīng)模型小鼠的影響,探討治療急性濕疹的藥效及可能的作用機制。 方法:1.昆明種健康雄性小鼠60只,18-22g,按體重分層隨機分為6組,每組10只。設(shè)模型對照組、基質(zhì)對照組、陽性對照組(地塞米松)、KG軟膏高、中、低劑量六個組,連續(xù)4天給藥,前三天每天給藥一次,每次只涂左耳,第四天左右耳均給藥,給藥兩次,末次給藥后1h后,滴20ul二甲苯于鼠左耳,1小時后將小鼠斷頸處死,沿耳廓基線部位剪下左右耳稱重,計算兩耳重量差異,即腫脹度。 2.昆明種健康雄性小鼠60只,18-22g,隨機分為6組,分別為正常對照組、模型對照組組、KG軟膏高劑量組、KG軟膏中劑量組、KG軟膏低劑量組,陽性對照組(地塞米松)。采用DNFB,腹部致敏、耳廓激發(fā)的方法建立小鼠遲發(fā)型超敏反應(yīng)模型。處死小鼠后,沿耳廓基線部位剪下雙耳,稱重,并計算腫脹度;左耳用10%福爾馬林溶液固定,HE染色,觀察耳廓充血、水腫及炎癥細(xì)胞浸潤的程度;并采用免疫組化的方法觀察KG軟膏對小鼠的耳組織中IFN-γ、IL-4、TNF-α、ICAM-1表達的影響。 結(jié)果:1.與模型組相比,高、中劑量組小鼠耳廓腫脹度均小于模型組,有統(tǒng)計學(xué)意義(P0.05或P0.01);低劑量組無明顯差異(P0.05),但有降低趨勢。表明KG軟膏對二甲苯所致小鼠急性耳腫脹有抑制作用,有抗急性炎癥作用。 2.與模型組相比,各給藥組小鼠耳廓腫脹度均小于模型組,有統(tǒng)計學(xué)差異(P0.01);KG軟膏各劑量組病理學(xué)評分均明顯降低,有統(tǒng)計學(xué)意義(P0.01);說明KG軟膏可降低DNFB激發(fā)后小鼠左耳的腫脹度,減輕左耳病理損傷,對DNFB所致小鼠遲發(fā)超敏反應(yīng)有抑制作用。 3.模型組與正常對照組比較,DNFB致敏小鼠耳組織中IFN-γ、IL-4、TNF-α、ICAM-1表達均明顯增高,有統(tǒng)計學(xué)意義(P0.05或P0.01);給藥各組IFN-γ、IL-4、TNF-α、ICAM-1的表達水平降低,有統(tǒng)計學(xué)意義(P0.05或P0.01),說明KG軟膏能抑制這些細(xì)胞因子的表達。 結(jié)論:1.KG軟膏能抑制二甲苯誘發(fā)的小鼠耳廓炎癥反應(yīng),2,4-二硝基氟苯誘發(fā)的小鼠遲發(fā)型超敏反應(yīng)、改善耳組織炎細(xì)胞浸潤情況、減輕細(xì)胞間水腫,表明KG軟膏有治療急性濕疹的作用。 2.KG軟膏治療急性濕疹作用機理部分與其抑制損傷局部IFN-γ、IL-4、TNF-α、ICAM-1等細(xì)胞因子的表達、調(diào)節(jié)T細(xì)胞亞群功能有關(guān)。
[Abstract]:Objective: to observe the effect of KG ointment on acute inflammation of auricle induced by xylene and delayed type hypersensitivity induced by DNFB in mice, and to explore the effect of KG ointment on acute eczema and its possible mechanism. Method 1: 1. Sixty healthy Kunming male mice were randomly divided into 6 groups according to their body weight, 10 rats in each group. Model control group, matrix control group, positive control group (high, middle and low dose of dexamethasone KG ointment) were given for 4 days. One hour after the last administration, the mice were killed by 20ul xylene dropping in the left ear for 1 hour. The left and right ears were cut off to weigh the right and left ears along the base line of the auricle, and the difference of the weight between the two ears was calculated, that is, the swelling degree. 2. Sixty Kunming healthy male mice were randomly divided into 6 groups: normal control group, model control group, high dose KG ointment group, low dose KG ointment group, and positive control group (dexamethasone). The delayed type hypersensitivity model of mice was established by DNFB, abdominal sensitization and auricle stimulation. After the mice were killed, the ears were cut off along the base line of the auricle to weigh and calculate the swelling degree, and the left ear was fixed with 10% formalin solution to fix the HE staining to observe the degree of congestion, edema and inflammatory cell infiltration of the auricle. The effect of KG ointment on the expression of ICAM-1 in mouse ear tissue was observed by immunohistochemical method. The result is 1: 1. Compared with the model group, the swelling degree of auricle in the middle dose group was lower than that in the model group (P0.05 or P0.01), while in the low dose group there was no significant difference (P 0.05), but there was a tendency to decrease. The results showed that KG ointment could inhibit acute ear swelling induced by xylene and inhibit acute inflammation in mice. 2. Compared with the model group, the swelling degree of auricle in each administration group was lower than that in the model group, and the pathological score of each dose of KG ointment was significantly lower than that of the model group, indicating that KG ointment could reduce the swelling degree of left ear after DNFB stimulation. The delayed hypersensitivity reaction induced by DNFB could be inhibited by alleviating the pathological injury of left ear. 3. The expression of ICAM-1 in the ears of DNFB-sensitized mice was significantly higher than that in the normal control group (P 0.05 or P 0.01), and the expression level of IFN- 緯 IL-4TNF- 偽 -ICAM-1 was lower in each group than that in the control group, and the expression of ICAM-1 was significantly higher in the model group than that in the normal control group, and the expression of ICAM-1 in the ear tissue of the DNFB sensitized mice was significantly lower than that in the control group. There was significant difference (P 0.05 or P 0.01), indicating that KG ointment could inhibit the expression of these cytokines. Conclusion 1. KG ointment can inhibit the auricular inflammation induced by xylene in mice, improve the infiltration of ear tissue cells, and alleviate the edema of the cells, and inhibit the delayed hypersensitivity of mice induced by dimethylbenzene and dinitrofluorobenzene. It indicated that KG ointment had the effect of treating acute eczema. The mechanism of 2.KG ointment in the treatment of acute eczema is partly related to its inhibition of the expression of cytokines such as IFN- 緯 -IL-4, TNF- 偽 and ICAM-1, and the regulation of the function of T cell subsets.
