龍葵堿抗腫瘤作用的研究進(jìn)展及臨床應(yīng)用

【摘要】:研究表明,目前來源于植物的天然成分成為當(dāng)前抗腫瘤藥物的研究熱點。龍葵堿是主要存在于龍葵全草、馬鈴薯及番茄中的甾體生物堿,除具有生物堿毒性作用外,還具有抗炎、抑菌、抗病毒的作用。近年來國內(nèi)外學(xué)者發(fā)現(xiàn),龍葵堿還具有明顯的抗腫瘤作用,使得眾多專家學(xué)者對其藥理作用及抗腫瘤機制進(jìn)行了深入的研究。本文對龍葵堿的化學(xué)組成和基本藥理作用,抗腫瘤作用機制和臨床應(yīng)用,特別是在惡性黑素瘤的作用及抗腫瘤機制等研究相關(guān)進(jìn)展進(jìn)行綜述,為其進(jìn)一步的科學(xué)研究和應(yīng)用提供科研思路,為皮膚惡性腫瘤,特別是在惡性黑素瘤治療尋求新的治療藥物方面提供依據(jù)。本研究基于“中國知網(wǎng)數(shù)據(jù)庫”、“萬方數(shù)據(jù)庫”及“PUBMED”等國內(nèi)外數(shù)據(jù)庫搜集的大量相關(guān)資料,篩選出近10年的有關(guān)龍葵堿抗腫瘤機制研究及臨床應(yīng)用等相關(guān)文獻(xiàn),并進(jìn)行閱讀、分析、整理而成。結(jié)果顯示:1)龍葵為一年生的草本植物,分布于全國各地,除具有生物堿毒性,能抗炎、抑菌、抗病毒,尚有抗腫瘤的作用。2)龍葵的主要藥理成分為甾體生物堿—龍葵堿,可以通過改變細(xì)胞膜蛋白的數(shù)量、功能、膜的通透性以及ATP酶、N-乙酰轉(zhuǎn)移酶的活性等多個環(huán)節(jié)來改變細(xì)胞膜結(jié)構(gòu)與功能。3)龍葵堿對人胰腺癌細(xì)胞系Panc-1細(xì)胞、人肝癌細(xì)胞系SMMC-7721細(xì)胞、胃癌細(xì)胞系NKM-45細(xì)胞等多種腫瘤細(xì)胞具有明顯的細(xì)胞毒性和抑制細(xì)胞增殖的作用,還可能通過信號轉(zhuǎn)導(dǎo)通路下調(diào)MMP-2/9、VEGF因子及鈣粘蛋白的表達(dá)水平。4)龍葵中藥方劑聯(lián)合放化療治療肺癌、胃癌、肝癌等多種腫瘤,均取得令人滿意的結(jié)果。5)龍葵堿在惡性黑素瘤細(xì)胞的實驗研究證實,其可能通過下調(diào)JNK通路和PI3K/Akt通路的信號傳導(dǎo)以及NF-κB因子的活性,直接或間接下調(diào)MMP-2/9的活性及蛋白的表達(dá)水平。綜上所述,龍葵堿對多種腫瘤細(xì)胞均具有生長抑制的作用及抗腫瘤機制;龍葵中藥方劑對臨床上多種腫瘤有減毒增效的作用;龍葵堿能夠抑制惡性黑素瘤細(xì)胞的遷移和侵襲能力,使細(xì)胞發(fā)生凋亡。此外,龍葵分布于全國各地,資源豐富,便于采集應(yīng)用于實驗及臨床研究。因此,龍葵堿在黑素瘤的治療方面有較大的開發(fā)和應(yīng)用前景。
[Abstract]:The research shows that the natural components from plants have become the research focus of anti-tumor drugs. Solanum nigrum alkaloids are steroidal alkaloids mainly found in Solanum nigrum L., potato and tomato. In addition to alkaloid toxicity, solanine also has anti-inflammatory, bacteriostatic and antiviral effects. In recent years, domestic and foreign scholars have found that solanine also has obvious anti-tumor effect, which makes many experts and scholars deeply study its pharmacological action and anti-tumor mechanism. In this paper, the chemical composition and basic pharmacological action of solanine, the mechanism of antitumor action and clinical application, especially in malignant melanoma and antitumor mechanism, were reviewed. To provide scientific research ideas for its further scientific research and application, and to provide scientific basis for the treatment of malignant skin tumors, especially in the treatment of malignant melanoma. Based on a large number of relevant data collected from domestic and foreign databases, such as "China knowledge Network Database", "Wanfang Database" and "PUBMED", this study screened out the relevant literatures on the mechanism of antitumor and clinical application of solanine in the last 10 years. And read, analyze, and organize. The results showed that: (1) Solanum nigrum is an annual herbaceous plant distributed all over the country. Besides alkaloid toxicity, anti-inflammatory, bacteriostatic, antiviral and anti-tumor effects, the main pharmacological component of Solanum nigrum is steroidal alkaloid-solanine. The cell membrane structure and function of human pancreatic cancer cell line Panc-1 can be changed by changing the number and function of cell membrane protein, membrane permeability and the activity of ATP enzyme N-acetyltransferase. Many kinds of tumor cells, such as human hepatoma cell line SMMC-7721 and gastric cancer cell line NKM-45, have obvious cytotoxicity and inhibit cell proliferation. It is also possible to down-regulate the expression level of MMP-2 / 9 + VEGF factor and cadherin through signal transduction pathway. 4) Solanum nigrum prescription combined with radiotherapy and chemotherapy in the treatment of lung cancer, gastric cancer, liver cancer and other tumors. The results showed that solanine might down-regulate the signal transduction of JNK pathway and PI3K/Akt pathway and the activity of NF- 魏 B factor in malignant melanoma cells. MMP-2/9 activity and protein expression were down-regulated directly or indirectly. To sum up, solanine has the effect of growth inhibition and anti-tumor mechanism on many kinds of tumor cells, the Chinese medicine prescription of Solanum nigrum has the effect of attenuating and enhancing many kinds of tumor in clinic. Solanine can inhibit the migration and invasion of malignant melanoma cells and induce apoptosis. In addition, Solanum nigrum is distributed all over the country, rich in resources, easy to collect and applied in experimental and clinical research. Therefore, solanine has a great prospect of development and application in the treatment of melanoma.
