發(fā)光二極管630nm紅光和460nm藍光照射對日本大耳白兔皮膚創(chuàng)面愈合的影響

【摘要】:目的觀察發(fā)光二極管(LED)630 nm紅光和460 nm藍光照射對日本大耳白兔皮膚創(chuàng)面愈合的影響。方法采用日本大耳白兔8只,建立兔背部皮膚創(chuàng)傷模型,每只兔子3個傷口,分別給予紅、藍光距離15 cm垂直照射(15 min/次)和自然愈合處理。照射至創(chuàng)傷后第21天,觀察各組創(chuàng)面愈合數(shù)目、愈合面積,計算創(chuàng)傷愈合面積百分比,比較兩種光源的治療作用;HE染色觀察新生組織結(jié)構(gòu);Masson染色觀察膠原纖維增生情況;免疫組織化學分析新生皮膚纖維細胞生長因子(FGF)、表皮生長因子(EGF)、內(nèi)皮細胞生長因子(CD31)、增殖細胞核抗原(Ki-67)及炎性細胞因子(CD68)的表達情況。結(jié)果紅光組、藍光組和對照組的愈合率分別為50.0%(4/8)、25.0%(2/8)和12.5%(1/8)。自造模后第12天起,紅光組傷口的愈合面積百分比均明顯高于藍光組及對照組(P0.05或P0.01)。建模后第21天,紅光組新生皮膚厚度為(2.95±0.34)mm,明顯高于對照組的(2.52±0.42)mm(F=3.182,P=0.016)。膠原纖維平均光密度為0.15±0.03,明顯高于藍光組的0.09±0.01(F=7.316,P=0.012)和對照組的0.07±0.01(F=7.316,P=0.003)。免疫組織化學檢測結(jié)果顯示,紅光組的EGF、FGF、CD31抗原、Ki-67表達較藍光組和對照組明顯增多,CD68較藍光組和對照組明顯減少(P0.05或P0.01)。結(jié)論 LED紅光照射可促進日本大耳白兔皮膚創(chuàng)面愈合,其可能是通過引起皮膚表皮細胞和血管內(nèi)皮細胞生長及纖維組織增生來促成的。
[Abstract]:Objective to observe the effects of red light (630 nm) and blue light (460 nm) on the healing of skin wounds in Japanese white rabbits.Methods 8 Japanese white rabbits were used to establish the model of skin injury on the back of rabbits. Each rabbit was treated with red and blue light for 15 min/ and natural healing.On the 21st day after irradiation, the number and area of wound healing were observed, and the percentage of wound healing area was calculated. The therapeutic effects of the two light sources were compared with HE staining to observe the proliferation of collagen fibers by Masson staining.The expression of fibroblast growth factor (FGFF), epidermal growth factor (EGFN), endothelial cell growth factor (CD31), proliferating cell nuclear antigen (Ki-67) and inflammatory cytokine (CD68) were analyzed by immunohistochemistry.Results the healing rates of red light group, blue light group and control group were 50. 0% and 25. 0% and 12. 5% 8 / 8 respectively.Since the 12th day after modeling, the percentage of wound healing area in red light group was significantly higher than that in blue light group and control group (P0.05 or P0.01).On the 21st day after modeling, the thickness of newborn skin in the red light group was 2.95 鹵0.34 mm, which was significantly higher than that in the control group (2.52 鹵0.42 mm).The average light density of collagen fibers was 0.15 鹵0.03, which was significantly higher than that of the blue light group (0.09 鹵0.01) and the control group (0.07 鹵0.01) and the control group (0.07 鹵0.01).The results of immunohistochemistry showed that the expression of Ki-67 in EGF- FGFGF-CD31 antigen in red light group was significantly higher than that in blue light group and control group. The expression of CD68 in red light group was significantly lower than that in blue light group and control group (P0.05 or P0.01).Conclusion LED red light irradiation can promote the healing of skin wounds in Japanese white rabbits, which may be induced by the growth of skin epidermal cells and vascular endothelial cells and the proliferation of fibrous tissue.
