【摘要】：目的建立SD大鼠外陰慢性單純性苔蘚(Lichen simplex chronicus,LSC)動物模型,研究蛋白酶激活受體2(protease activated receptor 2,PAR2)在模型大鼠外陰皮損中的表達,為LSC提供理想的動物模型。方法將70只雌性SD大鼠隨機分為A組(空白對照組,n=10)、B組(丙酮對照組,n=10)和C組(單純機械刺激對照組,n=10)、D組(實驗組,n=40)。A組不予以任何處理;B組每周3次涂丙酮溶液于大鼠外陰皮膚,共10周;C組每周3次對大鼠皮膚予以單純的機械刺激,共10周;D組予以0.5%二甲基苯并蒽(7,12-Dimethylbenzanthracene,DMBA)丙酮溶液聯(lián)合機械刺激每周3次誘導大鼠外陰皮膚,共10周。6、8、10、12、14周肉眼及鏡下觀察大鼠外陰皮膚的變化。免疫組織化學、Western blotting檢測A組和D組PAR2蛋白的表達,q RT-PCR檢測PAR2的m RNA的表達。結果 D組第8周有57.5%(23/40)的SD大鼠出現(xiàn)了LSC改變,第10周有95%(38/40)的大鼠外陰表現(xiàn)為LSC,至12周時,20%(8/40)的大鼠外陰都形成了乳頭狀新生物。丙酮及單純的機械刺激無導致LSC的作用。免疫組化及Western blotting結果顯示D組PAR2的蛋白表達量較A組明顯增加(P0.05);q RT-PCR結果顯示D組PAR2的m RNA的表達量較A組明顯增加(P0.05)。結論機械刺激聯(lián)合0.5%DMBA丙酮溶液可誘導雌性SD大鼠LSC,PAR2可能與疾病的發(fā)生及進展密切相關。
[Abstract]:Objective to establish an animal model of chronic lichen vulva simplex (Lichen simplex chronicusus) in SD rats, and to study the expression of 2(protease activated receptor 2 (PAR2) in vulvar lesions of the model rats, so as to provide an ideal animal model for LSC.Methods 70 female Sprague-Dawley rats were randomly divided into two groups: group A (control group), group B (acetone control group, n = 10) and group C (control group with mechanical stimulation), group D (n = 40). Group A was treated with acetone solution 3 times a week without any treatment.In rat vulva skin,The rats in group C were treated with mechanical stimulation three times a week for 10 weeks, and group D were treated with 0.5% dimethyl benzanthene 712-Dimethylbenzanthracene-DMBA solution combined with mechanical stimulation three times a week to induce vulvar skin.The changes of vulvar skin in rats were observed with naked eye and under microscope for 10 weeks.The expression of PAR2 protein in group A and D was detected by Western blotting. The expression of m RNA in PAR2 was detected by Q RT-PCR.Results in group D, 57.5% of SD rats had LSC changes at the 8th week, 95 / 38 / 40 of the rats' vulva showed LSC at the 10th week, and at 12 weeks, 20 / 40% of the vulva formed papillary neoplasm.Acetone and mechanical stimulation alone had no effect on LSC.Immunohistochemical and Western blotting results showed that the expression of PAR2 protein in group D was significantly higher than that in group A. The results showed that the expression of m RNA in PAR2 in group D was significantly higher than that in group A.Conclusion Mechanical stimulation combined with 0.5%DMBA acetone solution can induce LSC-PAR2 in female SD rats, which may be closely related to the occurrence and progression of the disease.
