祛斑方對紫外線照射后色素沉著動物模型病理形態(tài)學(xué)影響

【摘要】:目的:探討祛斑方對紫外線照射后色素沉著豚鼠模型的病理形態(tài)學(xué)影響。方法:采用中波紫外線照射白色雄性豚鼠復(fù)制黃褐斑實驗動物模型,用免疫組化法檢測祛斑方對皮膚黑素細(xì)胞的影響,并通過透射電鏡法觀察黑素小體的表達(dá)。結(jié)果:1黑色素陽性細(xì)胞的表達(dá):造模后,模型組與空白組比較黑色素陽性細(xì)胞的數(shù)量顯著增多(P0.05),與模型組比較,3個給藥組黑色素陽性細(xì)胞數(shù)量明顯下降(P0.05);祛斑方高劑量組顯著優(yōu)于西藥對照組(P0.05)。2免疫組化染色病理切片結(jié)果:空白組表皮可見到極少個黑素細(xì)胞,模型組表皮層增厚,表皮基底細(xì)胞和棘細(xì)胞層中可見多個黑素細(xì)胞,呈強(qiáng)陽性反應(yīng);祛斑方低、高劑量組表皮基底細(xì)胞和棘細(xì)胞層中可見一些散在分布的黑素細(xì)胞;西藥對照組表皮基底細(xì)胞和棘細(xì)胞層中可見數(shù)個散在分布的黑素細(xì)胞。3電子顯微鏡觀察結(jié)果:空白組黑色素細(xì)胞細(xì)胞漿豐富,含有極少量直徑小的黑素小體,模型組視野中可見表皮細(xì)胞中存在較多Ⅲ、Ⅳ期黑素小體,祛斑方高、低劑量組視野內(nèi)黑素小體、黑素生成均較少,西藥對照組細(xì)胞核上方可見數(shù)個黑素小體,直徑較小。結(jié)論:祛斑方對紫外線照射后色素沉著動物模型具有良好的治療作用,其機(jī)制可能是通過抑制黑素細(xì)胞的增殖,降低黑素細(xì)胞的功能達(dá)到的。
[Abstract]:Objective: to study the pathomorphological effect of Qubanfang on pigmented guinea pig model after ultraviolet irradiation.Methods: the experimental animal model of chloasma was established by ultraviolet B irradiation in white male guinea pigs. The effect of Qubanfang on melanocytes was detected by immunohistochemical method, and the expression of melanosome was observed by transmission electron microscope (TEM).Results the expression of melanin positive cells: after modeling,Compared with the blank group, the number of melanin positive cells in the model group was significantly higher than that in the blank group. Compared with the model group, the number of melanin positive cells in the three administration groups was significantly lower than that in the control group, and that in the high dose group was significantly better than that in the western medicine control group.The results of staining pathological sections showed that very few melanocytes could be seen in the epidermis of blank group.In the model group, many melanocytes were found in epidermal basal cell and spinous cell layer, and some scattered melanocytes were found in epidermal basal cell and spinous cell layer in high dose group.In the epidermal basal cell and spinous cell layer of the control group, the results of electron microscope observation showed that the melanocytes in the blank group were rich in cytoplasm and contained a very small number of melanosomes with small diameters.In the model group, there were more 鈪,
