【摘要】：目的研究蒺藜防白丸對(duì)黑素細(xì)胞(melanocytes,MC)的形態(tài)、增殖、酪氨酸酶活性及黑素合成的影響。方法采用倒置顯微鏡觀察細(xì)胞形態(tài),四甲基偶氮唑藍(lán)(MTT)法測(cè)定蒺藜防白丸對(duì)黑素細(xì)胞增殖的影響,多巴氧化法測(cè)定酪氨酸酶活性,NaOH法測(cè)定黑素含量。結(jié)果隨著蒺藜防白丸濃度的升高,黑素細(xì)胞樹突變粗短,且數(shù)量減少;低濃度(0.001~0.5mg/mL)的蒺藜防白丸對(duì)黑素細(xì)胞增殖影響不顯著(P0.05),高濃度時(shí)(1mg/mL和5mg/mL)抑制作用明顯(P0.05);低濃度的蒺藜防白丸可以提高酪氨酸酶活性,其中0.05mg/mL時(shí)酪氨酸酶活性最高(P0.05),然而當(dāng)蒺藜防白丸濃度繼續(xù)增大,則會(huì)對(duì)酪氨酸酶活性產(chǎn)生抑制作用;蒺藜防白丸對(duì)黑素細(xì)胞黑素合成也有類似的影響。結(jié)論蒺藜防白丸高濃度時(shí)抑制黑素細(xì)胞的增殖、酪氨酸酶活性及黑素合成;低濃度時(shí)促進(jìn)酪氨酸酶活性及黑素合成。
[Abstract]:Objective to study the anti white Tribulus terrestris Pill on melanocytes (melanocytes, MC) the effect of morphology, proliferation, tyrosinase activity and melanin synthesis method. Cell morphology was observed by inverted microscope, four methyl thiazolyl tetrazolium (MTT) method for the determination of anti white tribulus pill effect on melanocyte proliferation, tyrosinase activity was measured by DOPA oxidation method, NaOH method for the determination of melanin content. Results with increasing concentrations of anti white Tribulus terrestris pill, melanocytes mutation tree thick and short, and reduce the number of low concentration; (0.001~0.5mg/mL) the influence of anti white Tribulus terrestris Pill on melanocyte proliferation was not significant (P0.05), high concentration (1mg/mL and 5mg/mL) significantly inhibited (P0.05); low concentration of anti white tribulus pills can increase the activity of tyrosinase, tyrosinase activity and the 0.05mg/mL (P0.05), while the highest concentration of anti white tribulus pill continues to increase, it will cause the inhibition of tyrosinase activity; anti white tribulus Conclusion: pill has a similar effect on melanin synthesis. Conclusion Tribulus terrestris white pill can inhibit the proliferation of melanocytes, tyrosinase activity and melanin synthesis at high concentration, and promote tyrosinase activity and melanin synthesis at low concentration.
