【摘要】：目的：檢測(cè)特應(yīng)性皮炎(Atopic dermatitis, AD)患者外周血單個(gè)核細(xì)胞（Peripheral Blood Mononuclear Cells, PBMCs）中miR-155和miR-142-5p的表達(dá)情況，，為進(jìn)一步研究miR-155、miR-142-5p在特應(yīng)性皮炎發(fā)生、發(fā)展中的作用提供理論依據(jù)。材料與方法：收集8例AD患者及6例正常人外周靜脈血各5ml，人淋巴細(xì)胞分離液分離PBMCs，miRNA提取分離試劑盒提取分離PBMCs中的miRNA,采用實(shí)時(shí)熒光定量PCR法，檢測(cè)AD患者和正常人PBMCs中miR-155、miR-142-5p的表達(dá)水平。以U6snRNA為內(nèi)參，采用2-ΔΔCT法計(jì)算miR-155、miR-142-5p在A(yíng)D患者PBMCs中的相對(duì)表達(dá)量，分析AD的嚴(yán)重程度（SCORAD）評(píng)分與miR-155、miR-142-5p的表達(dá)水平之間的關(guān)系。用SPSS17.0統(tǒng)計(jì)軟件對(duì)實(shí)驗(yàn)數(shù)據(jù)進(jìn)行統(tǒng)計(jì)學(xué)分析，以α=0.05作為檢驗(yàn)標(biāo)準(zhǔn)。結(jié)果：1. miR-155、miR-142-5p在A(yíng)D患者PBMCs中均有表達(dá)。2.與正常對(duì)照組相比，miR-155、miR-142-5p在A(yíng)D患者PBMCs中的表達(dá)水平上調(diào)（p＜0.05）。3. miR-155的相對(duì)表達(dá)量與AD嚴(yán)重程度積分有相關(guān)傾向(r=0.695, p=0.056),而miR-142-p的相對(duì)表達(dá)量與其無(wú)相關(guān)性(r=0.283, p=0.497)。結(jié)論：1.特應(yīng)性皮炎患者PBMCs中存在miR-155、miR-142-5P，并可以用實(shí)時(shí)熒光定量PCR進(jìn)行檢測(cè)。2. miRNA-155、miRNA-142-5p在A(yíng)D患者PBMCs中表達(dá)上調(diào)，可能在A(yíng)D的發(fā)生、發(fā)展過(guò)程中發(fā)揮一定作用。3. miR-155的相對(duì)表達(dá)量與AD嚴(yán)重程度積分有相關(guān)傾向,而miR-142-p的相對(duì)表達(dá)量與其無(wú)相關(guān)性。
[Abstract]:Objective: to detect the expression of miR-155 and miR-142-5p in peripheral Blood Mononuclear cells (PBMCs) in patients with atopic dermatitis (AD), in order to further study the occurrence of miR-155 miR-142-5p in atopic dermatitis. Materials and methods: 5 ml of peripheral venous blood were collected from 8 patients with AD and 6 cases of normal controls. The isolation kit was used to extract miRNAs from peripheral blood of 8 patients with AD and 6 normal controls. Real time fluorescence quantitative PCR method was used to extract miRNAs from PBMCs. The expression of miR-155 miR-142-5p was detected in AD patients and normal subjects, and the relative expression of miR-155 miR-142-5p in PBMCs of AD patients was calculated by 2- 螖 CT using U6snRNA as internal control. The relationship between the severity of AD and the expression level of miR-155 miR-142-5p was analyzed. The experimental data were statistically analyzed by SPSS17.0 software. Results: 1. MiR-155 miR-142-5p was expressed in PBMCs of AD patients. The expression level of miR-155 miR-142-5p was up-regulated in PBMCs of AD patients compared with normal controls. The relative expression of miR-155 was significantly correlated with the score of AD severity. Conclusion: 1. MiR-155 miR-142-5P exists in PBMCs of patients with atopic dermatitis, and can be detected by real-time fluorescence quantitative PCR to detect upregulation of .2miRNA-155 miRNA-142-5p in PBMCs of AD patients. It may play a certain role in the occurrence and development of AD. The relative expression of miR-155 has a tendency to correlate with the score of AD severity, but the relative expression of miR-142-p has no correlation with AD.
