【摘要】： 目的 研究緊密連接蛋白JAM-1和claudin-7在尋常型銀屑病患者皮損組織中的表達(dá)水平,探討JAM-1蛋白和claudin-7蛋白在尋常型銀屑病中可能的發(fā)病機(jī)制。 方法 本課題選取25例尋常型銀屑病患者作為實(shí)驗(yàn)組,15例健康正常人作為對照組,所用研究方法如下： 1)采用免疫組化法(S-P法)檢測尋常型銀屑病患者皮損組織和正常對照者皮膚組織中JAM-1、claudin-7蛋白的表達(dá)水平； 2)應(yīng)用逆轉(zhuǎn)錄聚合酶鏈反應(yīng)法(RT-PCR法)檢測尋常型銀屑病患者皮損組織和正常對照者皮膚組織中JAM-1、claudin-7 mRNA的表達(dá)水平。 結(jié)果 1)尋常型銀屑病患者皮損組織中表皮層角質(zhì)形成細(xì)胞膜上JAM-1蛋白和claudin-7蛋白表達(dá)陰性,而正常對照組中JAM-1蛋白和claudin-7蛋白高表達(dá)； 2)與正常對照相比,尋常型銀屑病患者皮損組織中JAM-1mRNA表達(dá)顯著降低(銀屑病患者vs正常對照者：0.38±0.10 vs2.51±0.09,p=0.03),差異具有統(tǒng)計(jì)學(xué)意義；claudin-7 mRNA表達(dá)亦顯著下降(銀屑病患者vs正常對照者：0.27±0.12 vs 1.83±0.15,p=0.02),差異具有統(tǒng)計(jì)學(xué)意義。 結(jié)論 尋常型銀屑病皮損中JAM-1蛋白和claudin-7蛋白表達(dá)明顯降低,可能在銀屑病的發(fā)病中起重要作用。
[Abstract]:Purpose. To investigate the expression of JAM-1 and claudin-7 in psoriasis vulgaris and to explore the possible pathogenesis of JAM-1 and claudin-7 in psoriasis vulgaris. Method. In this study, 25 patients with psoriasis vulgaris were selected as experimental group and 15 healthy persons as control group. 1) the expression of JAM-1claudin-7 protein in the lesions of patients with psoriasis vulgaris and normal controls was detected by immunohistochemical staining (S-P method). 2) reverse transcriptase polymerase chain reaction (RT-PCR) was used to detect the expression of JAM-1claudin-7 mRNA in the lesions of psoriasis vulgaris and the normal controls. Results. 1) the expression of JAM-1 protein and claudin-7 protein in epidermal keratinocytes was negative in psoriasis vulgaris, but JAM-1 protein and claudin-7 protein were highly expressed in normal control group. 2) compared with normal control, The expression of JAM-1mRNA in lesions of psoriasis vulgaris patients was significantly lower than that in normal controls (P < 0. 38 鹵0. 10 vs2.51 鹵0. 09 vs2.51 鹵0. 03), and the expression of claudin-7 mRNA in psoriatic patients was significantly lower than that in normal controls (0. 27 鹵0. 12 vs 1. 83 鹵0. 15p0. 02). Conclusion. The expression of JAM-1 protein and claudin-7 protein in psoriasis vulgaris may play an important role in the pathogenesis of psoriasis.
