SIRT1過表達介導(dǎo)PPARγ-PGC1α-NRF2通路對肥胖癥大鼠肝臟脂肪沉積和變性及氧化應(yīng)激的調(diào)節(jié)作用(英文)

目的:探討沉默信息調(diào)節(jié)因子1(SIRT1)過表達介導(dǎo)PPARγ-PGC1α-NRF2通路對肥胖癥大鼠肝臟脂肪沉積和變性及氧化應(yīng)激的影響。方法:首先構(gòu)建SIRT1慢病毒載體(LV),然后將大鼠隨機分為4組:正常對照組(Control)、肥胖癥模型組(Obesity model組)、肥胖癥+SIRT1對照載體組(Obesity+LV組)和肥胖癥+SIRT過表達組(Obesity+LV-SIRT1組)進行后續(xù)實驗。采用卷尺測量肛鼻長并計算Lee’s指數(shù);羅氏血糖儀檢測血糖水平;全自動生化分析儀檢測總膽固醇(TC)、甘油三酯(TG)、低密度脂蛋白膽固醇(LDL-C)、高密度脂蛋白膽固醇(HDL-C)水平;蘇木精—伊紅染色法(hematoxylin-eosin staining,HE)和油紅O染色檢測肝臟脂肪沉積和變性;采用ELISA試劑盒檢測總超氧化物歧化酶(SOD)、乳酸脫氫酶(LDH)、丙二醛(MDA)和活性氧(ROS)含量;逆轉(zhuǎn)錄—聚合酶鏈反應(yīng)檢測過氧化物酶體增殖物激活受體γ(PPARγ)、PPARγ輔激活子1α(PGC1α)、核因子-E2相關(guān)因子2(Nrf2)m RNA水平;蛋白免疫印...
