吲哚丙烯酸調(diào)控腸道菌群和AhR激活阻抑強直性脊柱炎小鼠疾病進展的研究

背景&目的強直性脊柱炎(ankylosing spondylitis,AS)是一種自身免疫關(guān)節(jié)疾病,主要侵犯脊柱,并累及骶髂關(guān)節(jié)和周圍關(guān)節(jié),嚴重者可導(dǎo)致患者脊柱及四肢關(guān)節(jié)強直的終身畸形。然而,目前AS的致病機理仍不明確,常規(guī)治療效果仍不理想。最近研究發(fā)現(xiàn),吲哚丙烯酸作為芳香烴受體(aryl hydrocarbon receptor,AhR)的激活劑,在體外研究中被證實具有顯著的抗炎效果。AhR與腸道菌群密切相關(guān),且參與免疫炎癥的調(diào)控。巨噬細胞遷移抑制因子(macrophage migration inhibitory factor,MIF)受NF-κB信號調(diào)控,致病菌和促炎癥因子的刺激可上調(diào)其表達,且與AS的脊柱炎癥進展高度相關(guān)。那么,吲哚丙烯酸能否重塑腸道菌群結(jié)構(gòu)與功能,并通過激活A(yù)hR促進腸道屏障功能,抑制NF-κB/MIF炎癥通路,最終緩解AS的疾病進展?本研究目的在于:明確吲哚丙烯酸對AS小鼠模型的治療效果,闡明其背后的可能作用機制,為吲哚丙烯酸作為一種潛在治療AS的手段提供理論依據(jù)。方法30只雌性BALB/c小鼠隨機分成對照組,模型組,吲哚丙烯酸治療組。構(gòu)建蛋白聚糖誘...
