菊粉聯(lián)合游泳訓(xùn)練對(duì)代謝綜合征大鼠骨骼肌胰島素受體底物1及葡萄糖轉(zhuǎn)運(yùn)子4表達(dá)的影響

【摘要】:目的觀察菊粉聯(lián)合游泳訓(xùn)練對(duì)代謝綜合征(MS)大鼠骨骼肌胰島素受體底物1(IRS-1)、葡萄糖轉(zhuǎn)運(yùn)體子4(GluT4)表達(dá)的影響。方法高糖高脂高鹽喂養(yǎng)結(jié)合腹腔注射STZ建立MS大鼠模型,干預(yù)8周后檢測(cè)各組FBG、HbA_1c及FIns,計(jì)算HOMA-IR;HE染色法觀察各組肝臟病理變化;免疫印跡及免疫組織化學(xué)法檢測(cè)大鼠骨骼肌IRS-1、GluT4表達(dá)。結(jié)果與正常飲食對(duì)照組比較,模型組FBG、HbA_1c、FIns及HOMA-IR升高(P0.01),肝臟出現(xiàn)非酒精性脂肪性肝病(NAFLD)病理改變,骨骼肌IRS-1、GluT4表達(dá)降低(P0.01)。與模型組比較,菊粉聯(lián)合游泳訓(xùn)練組FBG、HbA_1c、FIns及HOMA-IR降低(P0.01),NAFLD減輕,骨骼肌IRS-1、GluT4表達(dá)升高(P0.01)。結(jié)論菊粉聯(lián)合游泳訓(xùn)練降低了MS大鼠FBG、HbA_1c及FIns水平,減輕了NAFLD病理改變,改善IR,其機(jī)制可能與上調(diào)骨骼肌IRS-1、GluT4表達(dá)有關(guān)。
[Abstract]:Objective to observe the effect of inulin combined with swimming training on the expression of insulin receptor substrate 1 (IRS-1) and glucose transporter 4 (GluT4) in skeletal muscle of (MS) rats with metabolic syndrome. Methods MS rat model was established by high glucose, high fat and high salt feeding combined with intraabdominal injection of STZ. After 8 weeks of intervention, FBG,HbA_1c and FIns, were detected by HOMA-IR;HE staining, and the expression of IRS-1,GluT4 in skeletal muscle of rats was detected by immunoblotting and immunohistochemistry. Results compared with the normal diet control group, FBG,HbA_1c,FIns and HOMA-IR in the model group were increased (P01), (NAFLD) pathological changes were found in the liver, and the expression of IRS-1,GluT4 in skeletal muscle was decreased (P01). Compared with the model group, FBG,HbA_1c,FIns and HOMA-IR in the inulin combined swimming training group decreased (P 0.01), NAFLD) and the expression of IRS-1,GluT4 in skeletal muscle increased (P 0.01). Conclusion inulin combined swimming training can decrease the levels of FBG,HbA_1c and FIns and alleviate the pathological changes of NAFLD in MS rats. The mechanism of improving IR, may be related to the up-regulation of IRS-1,GluT4 expression in skeletal muscle.
