茶多酚對果糖誘導(dǎo)的高尿酸血癥大鼠血尿酸水平的影響及機制

【摘要】:目的:觀察茶多酚(green tea polyphenols,GTP)對果糖誘導(dǎo)的高尿酸血癥(hyperuricemia,HUA)大鼠血尿酸水平的影響,并從調(diào)節(jié)尿酸產(chǎn)生和排泄途徑探討其藥理機制。方法:連續(xù)給予大鼠10 g/100 mL果糖溶液8周,其中5~8周分別連續(xù)灌胃別嘌呤醇(4 mg/(kg·d))或GTP(75、150、300 mg/(kg·d)),每天1次。磷鎢酸法檢測血尿酸水平,比色法和Western blot法分別檢測肝臟黃嘌呤氧化酶(xanthine oxidase,XOD)活力及其表達水平,免疫組織化學(xué)染色法檢測腎臟中尿酸鹽轉(zhuǎn)運子(urate transporter,UAT)、尿酸鹽陰離子轉(zhuǎn)運子(urate-anion transporter,URAT)1和葡萄糖轉(zhuǎn)運子(glucose transporter,GLUT)9的表達水平。結(jié)果:150、300 mg/(kg·d)GTP顯著降低了果糖誘導(dǎo)的HUA大鼠血尿酸水平(P0.05,P0.01)。同時,150、300 mg/(kg·d)GTP顯著降低了HUA大鼠肝臟XOD活力和表達水平(P0.05,P0.01),顯著增強了腎臟UAT表達水平和降低了腎臟URAT1和GLUT9的表達水平(P0.05,P0.01)。結(jié)論:150、300 mg/(kg·d)GTP可有效降低果糖誘導(dǎo)的HUA大鼠血尿酸水平,其機制與減少尿酸產(chǎn)生和促進尿酸排泄密切相關(guān)。
