維生素C治療百草枯中毒大鼠的量效關(guān)系
[Abstract]:Objective: to investigate the efficacy of vitamin C in the treatment of paraquat (paraquat,PQ) poisoning in rats. Methods: 40 SpragueDawley (SD) rats were randomly divided into three groups: control group, intoxication group, vitamin C 1 group and C 2 group, with 10 rats in each group. The rat model of PQ poisoning SD was established by intragastric infusion of 150 mg/kg PQ,. In the poisoning group, 30 mg/kg methylprednisolone was injected intraperitoneally with 2.5 mg/kg cyclophosphamide. Two concentrations of vitamin C (5500 mg/kg) were given to vitamin C group and C2 group, diluted into 5 mL 路kg ~ (- 1) with saline and injected intravenously into femoral vein, while the control group was treated with normal saline only. Malondialdehyde (MDA) in plasma and function of liver and kidney were measured at 36 h after the experiment, and blood gas analysis was performed. Lung tissue was sacrificed after blood collection for pathological section and wet-dry weight ratio (WW / W). The expression of glutathione peroxidase (Bcl-2) in liver tissue was detected by glutathione peroxidase,GSH-PX), Western blot. Results: MDA in vitamin C1 group was lower than that in poisoning group, and GSH-PX was higher than that in poisoning group. The liver and kidney function of vitamin C1 group was significantly improved than that of poisoning group (all P0.01). The MDA of vitamin C _ 2 group was higher than that of poisoning group, and the liver and kidney function of vitamin C _ 2 group was worse than that of poisoning group (P 0.01). The expression of Bcl-2 in the poisoning group was lower than that in the control group, the expression of Bcl-2 in the vitamin C1 group was higher than that in the poisoning group, and the expression of Bcl-2 in the vitamin C2 group was lower than that in the poisoning group (P0.01). However, there was no significant difference in lung function, wet-dry weight ratio and pathology between vitamin C _ 1 and C _ 2 groups (P 0.05). Conclusion: low dose vitamin C may have partial protective effect on liver and kidney function of PQ poisoned rats by anti-oxidation and inhibiting apoptosis induced by PQ, while high dose vitamin C can promote oxidation. At the same time, vitamin C did not show a protective effect on the lung after PQ poisoning.
