【摘要】：目的:篩選類(lèi)風(fēng)濕關(guān)節(jié)炎(RA)患者外周血與關(guān)節(jié)炎大鼠破骨細(xì)胞的miRNAs異常表達(dá)譜,分析RA的特征性miRNAs及其功能。方法:采用miRNAs芯片,篩選RA患者與正常人外周血、共培養(yǎng)誘生的關(guān)節(jié)炎大鼠破骨細(xì)胞與正�；|(zhì)細(xì)胞的miRNAs表達(dá)譜的差異;Real time PCR對(duì)芯片結(jié)果進(jìn)行驗(yàn)證;生物信息學(xué)方法對(duì)關(guān)鍵miRNA進(jìn)行功能分析。結(jié)果:與正常人相比,RA患者外周血中具有差異表達(dá)的miRNAs有189個(gè);與單核細(xì)胞比較,破骨細(xì)胞中具有差異表達(dá)的miRNAs有211個(gè),其中miR-15b-5p等10個(gè)miRNAs在RA患者外周血及大鼠破骨細(xì)胞中均異常表達(dá)。Real time PCR驗(yàn)證結(jié)果與芯片檢測(cè)結(jié)果具有一致性。生物信息學(xué)分析結(jié)果顯示特征性miRNA的靶基因顯著富集在VEGF,MAPK信號(hào)通路等信號(hào)通路。結(jié)論:部分miRNAs在RA患者外周血及關(guān)節(jié)炎大鼠破骨細(xì)胞中的異常表達(dá)具有一致性,其可能作為RA的特征性miRNAs,通過(guò)調(diào)控相關(guān)信號(hào)通路干預(yù)破骨細(xì)胞分化等病理過(guò)程。
[Abstract]:Aim: to screen the abnormal miRNAs expression profiles of osteoclasts in peripheral blood and osteoclasts of patients with rheumatoid arthritis (RA) and to analyze the characteristic miRNAs and its function of RA. Methods: the miRNAs expression profiles of osteoclasts and normal stromal cells in osteoclasts of induced arthritis rats were cocultured by miRNAs microarray, and the results were verified by; Real time PCR. Bioinformatics method was used to analyze the function of key miRNA. Results: there were 189 miRNAs in peripheral blood of RA patients. Compared with monocytes, there were 211 miRNAs differentially expressed in osteoclasts, of which 10 miRNAs, including miR-15b-5p, expressed. Real time PCR in peripheral blood of RA patients and in osteoclasts of rats. The results were consistent with the results of microarray detection. Bioinformatics analysis showed that the target genes of characteristic miRNA were significantly enriched in signal pathways such as VEGF,MAPK signaling pathway. Conclusion: the abnormal expression of partial miRNAs in peripheral blood of RA patients and osteoclasts in arthritis rats is consistent. It may be the characteristic miRNAs, of RA to interfere with osteoclast differentiation and other pathological processes by regulating related signaling pathways.
