羅格列酮對OLETF大鼠肝臟組織病理變化及白細(xì)胞介素-17、Bcl-xl表達(dá)的影響

【摘要】:目的探討羅格列酮(RGZ)對2型糖尿病(T2DM)OLETF大鼠肝臟組織病理變化及白細(xì)胞介素(IL)-17、Bcl-xl的影響。方法取OLETF雄性T2DM大鼠16只,隨機分為T2DM組和RGZ組,正常對照組(NC)組為LETO鼠,每組8只。給藥12 w后同時采集肝臟組織,進行HE、Mass、PAS染色觀察組織學(xué)改變,免疫組織化學(xué)染色觀察IL-17和Bcl-xl陽性細(xì)胞。結(jié)果 T2DM組大鼠肝小葉結(jié)構(gòu)紊亂,可見大量膠原纖維。肝細(xì)胞脂肪空泡形成、糖原沉積減少、細(xì)胞凋亡增加,Bcl-xl蛋白表達(dá)降低、炎癥因子IL-17增加。RGZ組較T2DM組肝細(xì)胞損傷明顯減輕。結(jié)論 T2DM可致肝臟組織學(xué)發(fā)生損傷,而羅格列酮可減輕這種損傷的發(fā)生。
[Abstract]:Objective to investigate the effect of rosiglitazone (RGZ) on hepatic histopathological changes and interleukin-17 (IL) Bcl-xl in rats with type 2 diabetes mellitus (T2DM). Methods Sixteen OLETF male T2DM rats were randomly divided into T2DM group and RGZ group. The normal control group (NC) group was LETO rats with 8 rats in each group. After 12 weeks of administration, liver tissues were collected at the same time, HE,Mass,PAS staining was used to observe the histological changes, and IL-17 and Bcl-xl positive cells were observed by immunohistochemical staining. Results in T2DM group, the hepatic lobular structure was disordered and a large number of collagen fibers were observed. Hepatocyte fat vacuole formation, glycogen deposition decreased, apoptosis increased, Bcl-xl protein expression decreased, and inflammatory factor IL-17 increased. The damage of hepatocytes in RGZ group was significantly reduced than that in T2DM group. Conclusion T2DM can induce liver histologic injury, and rosiglitazone can reduce it.
