無癥狀高尿酸血癥大鼠血液流變學(xué)和氧化應(yīng)激

【摘要】:目的從血液流變學(xué)的角度對(duì)長期無癥狀高尿酸血癥開展系統(tǒng)研究,為該癥的臨床處理提供參考依據(jù)。方法將20只SD大鼠隨機(jī)平均分為空白對(duì)照組和模型組。通過對(duì)大鼠腹腔注射250 mg/(kg·d)氧嗪酸鉀誘導(dǎo)8周無癥狀高尿酸血癥模型,采血測(cè)量尿酸水平、血液流變學(xué)指標(biāo)、氧化和抗氧化指標(biāo)。結(jié)果模型組大鼠紅細(xì)胞的聚集指數(shù)、破碎率、血清黃嘌呤氧化酶活性(XOD)、血漿纖維蛋白原、血液黏度顯著升高,紅細(xì)胞的取向指數(shù)、電泳率、血清超氧化物歧化酶活性(SOD)、活化部分凝血酶時(shí)間(APTT)和凝血酶原時(shí)間(PT)顯著降低。結(jié)論無癥狀高尿酸血癥水平下機(jī)體氧化應(yīng)激增強(qiáng),使大鼠紅細(xì)胞流變特性發(fā)生不良變化,血液處于高黏高凝狀態(tài)。研究結(jié)果提示臨床診治中應(yīng)對(duì)無癥狀高尿酸血癥正確認(rèn)識(shí)并及時(shí)干預(yù)。
[Abstract]:Objective to systematically study chronic asymptomatic hyperuricemia from the perspective of hemorheology, and to provide reference for clinical management of the disease. Methods 20 SD rats were randomly divided into blank control group and model group. The model of asymptomatic hyperuricemia was induced by intraperitoneal injection of potassium oxazinate for 8 weeks. The levels of uric acid, hemorheology, oxidation and antioxidation were measured. Results in the model group, the aggregation index, fragmentation rate, serum xanthine oxidase activity, plasma fibrinogen, blood viscosity, erythrocyte orientation index and electrophoretic rate were significantly increased. The activity of serum superoxide dismutase (SOD),) activated partial thrombin time (APTT) and prothrombin time (PT) significantly decreased. Conclusion oxidative stress is enhanced under asymptomatic hyperuricemia, which results in adverse changes of erythrocyte rheological characteristics and hyperviscosity and hypercoagulability. The results suggest that the correct understanding of asymptomatic hyperuricemia and timely intervention should be taken in clinical diagnosis and treatment.
